MTHFR基因低表达和环境危险因素与食管癌发生的关系

摘    要:[目的]了解亚甲基四氢叶酸还原酶基因(MTHFR基因)的表达与食管癌发生的关系,探讨食管癌相关环境危险因素与MTHFR基因表达水平之间的交互作用。[方法]收集97例淮安地区食管癌新发病例的食管癌组织及远端癌旁组织,采用实时荧光定量RT-PCR方法定量分析食管癌组织与癌旁组织中MTHFR基因的表达水平,同时运用单纯病例研究分析MTHFR mRNA水平与环境危险因素之间的交互作用。[结果]MTHFR mRNA表达水平在食管癌组织显著低于癌旁组织,与癌旁组织相比,癌组织的MTHFR mRNA表达水平下调了48%,条件Logistic回归分析表明MTHFR mRNA表达下调与食管组织癌变有统计学关联(OR=I.20,95%CI:1.05~1.37);饮茶习惯与MTHFR mRNA低表达之间存在拮抗作用,且有剂量一反应关系。饮酒与MTHFR mRNA低表达之间的协同作用无统计学意义。[结论]MTHFR mRNA表达水平可反映食管组织叶酸代谢的能力,其下调与食管癌发生有关;饮茶可拮抗MTHFR mRNA表达缺陷对食管癌发病的影响。
