肝癌组织中HGFAC表达与DNA甲基化及预后的相关性

目的:探讨肝癌手术患者癌组织中肝细胞生长因子激活因子(HGFAC)表达与DNA甲基化及预后的相关性研究。方法:选取延安大学附属医院普通外科行手术治疗的肝癌患者46例为肝癌组,肝外伤手术治疗患者20例为对照组。两组手术前后分别采用酶联免疫定量法、蛋白质免疫印迹法、免疫组织化学法检测患者手术后肝组织中HGFAC表达,采用Kaplan-Meier法计算生存率并进行生存分析。结果:HGFAC表达与血清甲胎蛋白、总胆红素、谷丙转氨酶指标具有相关性(P<0.05);肝癌组术后2、6、12、24 h各时间段血清HGFAC水平均低于对照组(P<0.05);免疫组织化学结果显示,对照组中HGFAC的表达水平明显高于肝癌组(P<0.05);HGFAC低表达患者的生存率明显低于HGFAC高表达患者,差异有统计学意义(P=0.003)。结论:HGFAC在肝癌组织中的表达降低与不良生存结局相关,DNA高甲基化导致HGFAC表达下降,HGFAC可能是预测肝癌预后的一种生物标志物。

Correlation between HGFAC expression and DNA methylation and prognosis in liver cancer

Objective:To explore the correlation between the expression of hepatocyte growth factor activator in tissues of patients undergoing surgery for liver cancer and DNA methylation and prognosis.Methods:Two groups of patients before and after surgery the research subjects selected 46 patients with liver cancer who underwent surgical treatment in the General Surgery Hospital of Yan'an University as the liver cancer group,and the control group selected 20 patients with liver trauma surgery as the control group.Enzyme-linked immunosorbent assay,western blotting,and immunohistochemistry were used to detect the expression of HGFAC in the liver tissues of patients after surgery.The Kaplan-Meier method was used to calculate the survival rate and perform survival analysis.Results:The expression of HGFAC were correlated with the indexes of serum alpha-fetoprotein,total bilirubin,and alanine glutamate aminotransferase(P<0.05);Serum HGFAC levels in the liver cancer group at 2,6,12,and 24 hours after surgery were significantly lower than those in the control group(P<0.05);In the expression of HGFAC in the two groups of liver tissue staining,the expression level of HGFAC in the control group was significantly higher than that in the liver cancer group(P<0.05);the survival rate of patients with low HGFAC expression was significantly lower than that of patients with high HGFAC expression Rate,there is a significant difference with statistical significance(P=0.003).Conclusion:The expression of HGFAC in serum is significantly reduced and the level of DNA methylation protein in plasma is high,indicating that the expression of HGFAC may be controlled by DNA methylation.In addition,the decrease in HGFAC is related to the significantly shorter survival time of patients with liver cancer.
