SD大鼠血常规指标正常参考值范围探讨

目的建立四川省疾病预防控制中心毒理所实验室30d喂养试验中雌雄远交群（Sprague-DawleySD）大鼠6项血常规指标正常参考值范围。方法初离乳SD大鼠220只,分11批,单笼喂饲30d后,禁食16h,不禁水,股静脉取血,EDTA-2K作抗凝剂,进行白细胞计数、红细胞计数、血红蛋白含量、白细胞3分类等6项指标测定。各项指标分雌雄进行正态性检验,符合正态分布用x^-±1.96s;雌雄大鼠指标两两比较用t检验。结果该验室雌雄SD大鼠6项血常规指标正常参考值范围分别为：白细胞计数2.92～11.15×10^9/L、2.08～11.78×10^9/L;红细胞计数6.16～7.97×10^12/L、6.04～8.33×10^12/L;血红蛋白含量124.23～163.19g/L、123.81～166.04g/L;淋巴细胞62.76%～90.19%、57.56～91.79%;中性细胞8.83%～31.17%、6.54%～36.69%;中值细胞0～7.11%、0.48%～6.93%。结论该实验室已建立雌雄大鼠血常规指标正常参考值范围,对其安全性毒理学评价有重要意义。

Range of Normal Reference Value of Routine Blood Examination Index for SD Rat

Object To determine the normal reference value range of 6 indices of routine blood examination for male and female SD rats in 30 d oral toxicity test in our laboratory. Method A total of 220 weanling SD rats of 11 fed-batchs were fed in cage separately for 30 days and blood were taken from femoral vein with EDTA-2K anti-freezing after being fasted for 16 hours （water was provided）. WBC, RBC, HGB and 3 indices of classification of leukocyte were detected. The test of normality was conducted for male and female rats. x ^-± 1.96s was used for indices with normality and t test was used for paired comparison of the indices of male and female rats. Results The range of normal reference value of 6. indices of blood routine examination for male and female SD rats in our laboratory were as follows： WBC 2. 92-11.15 ×10^9/L and 2. 08-11.78 × 10^9/L; RBC 6. 16-7. 97 × 10^12 /L and 6. 04-8.33 × 10^12/L; HGB 124. 23 -163. 19 g/L and 123.81-166. 04 g/L; LPR 62. 76-90.-19% and 57.56-91.79% ; GPR8. 83-31.17% and 6. 54-36. 69% ; MPR 0-7. 11% and 0. 48-6. 93%. Conclusion Determination of the normal reference value range of routine blood examination indices for male and female SD rats in laboratory is important for toxicological assessment on safety.
