摘    要:目的 模拟相血糖控制不佳出现的糖浓度波动,研究波动性细胞外高糖浓度(FEHG)对肾小球系膜细胞(MsC)增殖及其纤维连接蛋白合成的影响。方法 利用四甲基偶氮唑蓝(MTT)比色法和竞争抑制ELISA法动态观察不同葡萄糖浓度及波动性高糖浓度对MsC增殖和FN合成的作用。结果 高糖浓度抑制MsC增殖,此抑制作用和糖浓度成正相关,同时刺激MsC的FN合成增加;

Abstract:Objective: In Order to observe the effects of hyperlgycemia on Mesangial cell(MsC). The effects of fluctuating extracellular high glucose(FEHG) on the proliferation and fibronectin (Fn) synthesis were studied. Methods:The Fn level in supernatant of culturedd MsC and MsC Proliferation were assessed by means of competitive inhibiting ELISA method and MTT colormetric method respectively. Results: High glucose levls inhibited the MsC proliferation and FEHG had the same inhibition effect. the Fn synthesis in FEHG was more higher than that in high glucose level. Conclusion: Fluctuating changes in extracellular glucose concentration may play an important pathological role for FN synthesis of MsC in patients with diabetes mellitus treated inadequatly.
