精浆松弛素对人精子运动及其相关参数的影响

摘    要:目前一些学者认为内源性松弛素与精子运动率有关,但报道不尽一致。本实验采用RLx抗体中和精液RLx的方法,观察内源性RLx对精子运动率的影响。收集22例精液标本,13例为正常运动率精子,9例为低活动率精子,待液化后进行精液常规分析,作各项运动参数的测定,然后每例标本取精液0.38ml加入RLx抗血清(滴度为1:6000)0.02ml作为实验组,另取精液0.38ml加入正常血清0.02ml作为对照组,置37℃水浴孵育.分别在15min、30min、60min时再次测定精子运动的各项参数并进行对比。结果显示:实验组除2例正常运动率精子变化不明显外,其余均呈运动率的明显下降;其中尤以精子前向运动率的抑制最为显著。实验证明:内源性RLx在维持精子活动能力.特别是前向运动过程中起着重要作用。

Abstract:To clarify the role of endogenrous relaxin in sperm motility,relaxin activity of human spermatozoa was neutralized in vitro,then sperm motility and relative motility parameters analyzed to detect some changes in response to the immunoneutralization. 22 semen specimens were collected from infertility clinic, 13 of which were of normal motility sperm,and the other 9 were of low motility sperm. After liquefaction,0. 76 ml was taken from each semen specimen and divided ran domly into two parts. 0. 38 ml semen to which was added 0. 02 ml anti-porcine RLx serum (titer 1: 6000) was used as experimental group and the other 0. 38 ml semen was with addition of 0. 02 ml serum only as control group. All samples were incubated at 37℃. Computerized semen analysis and sperm motility characteristics were performed with Hamilton-Thorn 2000 Motility Analyzer to detect sperm motility rate (% ), progressive motility rate (% ),path velocity (micro/sec) and velocity (0~4) at the time of 0 min, 15 min, 30 min and 60 min respectively. The data were compared by T-test with dependent samples program. The results showed that sperm motility declined significantly after incubation with anti-RLx serum except for two cases. Sperm progressive motility declined most significantly. This experiment revealed that endogenous RLx played an important role in the physiological process of maintaining sperm progressive motility.
