山药多糖对糖尿病肾病大鼠肾功能及肾MCP-1表达的影响

摘    要:目的探讨山药多糖(Chineseyampolysaccharides,CYP)对糖尿病肾病(Diabeticnephropathy,DN)大鼠肾功能及肾脏单核细胞趋化蛋白-1(Monocytechemptacticprotein-1,MCP-1)mRNA表达的影响。方法70只wistar大鼠随机分为对照组和糖尿病模型组,后者以链脲佐菌素(sTZ)60mg·kg-’诱导糖尿病肾病大鼠模型,成模大鼠按干预方法分为糖尿病肾病模型组(DN组)、氯沙坦(LsT组)和CYP低、中、高剂量(cYP_L、CYP-M、cYP-H)5组,给药后6周及8周末,采用放射性免疫法观察大鼠24h尿蛋白水平;采用自动生化仪检测血清中血糖(GLU)、尿素氮(BUN)、肌酐(SCr)水平;采用Real—timePCR法观察肾组织CMP-1 mRNA表达。结果与DN组比较,LST组及CYP中、高剂量组24h尿蛋白、血糖、尿素氮、肌酐水平均显著降低;同时下调大鼠肾组织CMP-1 mRNA的表达。结论CYP能降低糖尿病肾病大鼠尿蛋白、尿素氮和肌酐水平,并下调肾组织CMP-1 mRNA的表达,提示早期应用CYP有预防和减轻肾损伤的可能。

Abstract:Objective To investigate the effects of Chinese yam polysaccharides on renal function and CMP-1 mRNA expression in diabetic nephropathy rats. Methods Seventy adult male Wistar rats were randomly divided into normal control group(NC)(10 rats)and diabetic nephropathy group(DN)(60 rats). DN rats were induced by peritoneal injection of streptozocin (SZT)of 60 mg'kg-l,then DN rats were divided into five groups:DN,losartan (I_ST),CYP-L,CYP-M and CYP-H,respectively. After 6 weeks and 8 weeks,The levels of 24 hours urine albumin proteins were detected by radioimmunoassay;that of plasma glucose,urea nitrogen,creatinine were detected by automatic biochemical analyzer,the CMP-1 mRNA expression of renal were detected by Real-time PCR. Results Diabetes nephropathy model were successfully induced. After treatment with CYP,the levels of 24 hours urine albumin protein of rats were significantly decreased and the levels of GLU, BUN and SCr were significantly decreased by CYP,and renal expression of CMP-1 mRNA were significantly down-regulated. Conclusions CYP could decrease the levels of urine albumin protein, BUN and SCr in DN rats,and down-regulate the CMP-1 mRNA expression in DN rats. This study shows that early application of CYP could effectively prevent the renal injury.
