摘    要:目的]测定朝鲜大黄中大黄酸的含量.方法]采用反相高效液相色谱法,色谱柱为VP-ODS色谱柱(4.6 mm×150 mm,5μm),以1 mL/L甲醇-磷酸(85∶15)为流动相,流动速度为1.0 mL/min,检测波长为254 nm.结果]大黄酸进样量在0.08~0.80μg范围内,其峰面积积分值与质量浓度之间呈良好的线性关系(r=0.999 8),平均回收率为99.58%(n=6),RSD为2.85%.药用大黄及朝鲜大黄中大黄酸含量分别为2.562 8,12.088 8 mg/g.结论]反相高效液相色谱法测定朝鲜大黄中大黄酸含量的方法简便、准确,可用于朝鲜大黄中大黄酸含量的测定;朝鲜大黄中大黄酸含量高于药用大黄.

Abstract:OBJECTIVE To appraise the quality of Rheum coreanum Nakai. METHODS RP-HPLC was carried out on VP-ODS column (4.6 mm×150 mm, 5μm) with methanol (1 mL/L) phosphotic acid (85 :15) as mobile phase. The flow rate was 1.0 mL/min and the UV detection was 254 nm. RESULTS The calibration curve was liner in the range of 0.08-0.80μgg (r= 0 . 999 8). The average recovery was 99.58o//00 (n=6) with the RSD of 2. 850%x. The content of rhein in Rheum officinale and R. coreananum was 2. 562 8 , 12. 088 8 mg/g. CONCLUSION The method appears to be simple, accurate, reproducible and can be used to determine the content of rhein in Rheum coreanum. The rhein content in Rheum coreanum is higher than that in Rh. officinale.
