| 摘 要： | 目的 构建人肝细胞生长因子 (hHGF)的原核表达载体。方法 利用基因重组技术 ,将人肝细胞生长因子cDNA全长序列 ,定向克隆于表达型载体PUC18质粒的多克隆位点中 ,限制性内切酶酶切鉴定。结果 重组PUC18-HGF原核表达载体经限制性内切酶电泳分析 ,与理论值相符。结论 本实验成功将人肝细胞生长因子cDNA全长序列插入表达型载体PUC18多克隆位点BamHI和Xbal限制性内切酶位点之间 ,为通过基因工程获得外源性HGF提供实验依据

| Abstract: | Objective:To construct prokaryotic expression plasmid containing hepatocyte growth factor. Methods:To insert human hepapocyte growth factor cDNA in multiple cloning site of the prokaryotic plasmid PUC18 by DNA recombinant technique and identify the sequence with restriction endonuclease digestion.Results:The sequence of recombinant expression vector PUC18-HGF digested with the enzyme corresponds to the expectation.Conclusions: The prokaryotic expression plasmid PUC18-HGF has been reconstructed successfully. It provides the experimental base for producing ectogeneous HGF by genetic engineering. |
