头体针对局灶性脑缺血大鼠脑组织CREB表达的影响

摘    要:目的:探讨头体针对局灶性脑缺血大鼠顶叶皮质cAMP反应元件结合蛋白(CREB)和磷酸化CREB(p-CREB)表达的影响。方法:SD雄性大鼠90只,随机分为正常组、模型组及针刺组各30只。模型组及针刺组大鼠采用中脑动脉闭塞(MCAO)法成功制作为局灶性脑缺血模型。针刺组于大鼠清醒后约1 h即平刺百会穴及点刺水沟穴,并连接电刺激;直刺双侧三阴交和丰隆穴,留针30 min,每日1次。治疗15 d后3组均采用神经行为学评分;应用实时定量PCR、Western Blotting和图像分析方法检测大鼠缺血侧顶叶皮质CREB和pCREB的表达。结果:造模后1~2 h及15d时神经症状行为评分,模型组及针刺组均明显高于正常组(P〈0.01);针刺组在治疗15 d时较治疗前及模型组有明显降低(P〈0.05),模型组则变化不明显。脑组织CREB的mRNA表达,模型组及针刺组均较正常组明显降低,针刺组明显高于模型组(P〈0.05)。结论:头体针能明显上调局灶性脑缺血大鼠顶叶皮质CREB和pCREB的表达,对缺血神经元有保护作用。
