EGFL7在人脑胶质瘤中的表达及其意义

目的 探讨EGFL7基因在人脑胶质瘤组织中的表达及其意义。方法 收集36例新鲜胶质瘤标本，其中包括21例低度恶性(Ⅰ~Ⅱ级)及15例高度恶性者 (Ⅲ~Ⅳ级)，用RT-PCR检测其EGFL7 mRNA的表达。另收集45例胶质瘤石蜡标本，其中包括24例低度恶性(Ⅰ~Ⅱ级)及21例高度恶性者(Ⅲ~Ⅳ级)，用免疫组织化学检测EGFL7蛋白的表达情况。并分析EGFL7蛋白表达与胶质瘤病理分级的关系。结果EGFL7 mRNA及其蛋白在人脑胶质瘤中存在表达，EGFL7蛋白表达主要定位于肿瘤细胞及血管内皮细胞的胞质。其表达与肿瘤的恶性程度有明显的相关性(t＝4.399，P<0.01)；EGFL7在正常脑组织中没有表达。结论人脑胶质瘤中肿瘤细胞及其血管内皮细胞EGFL7均呈增高表达并且与其肿瘤恶性程度具有明显相关性，提示EGFL7在胶质瘤的发生发展中发挥重要作用。

Expression of EGFL7 in Human Gliomas and Its Correlation with Glioma Grade

ObjectiveTo investigate the expression of EGFL7 in human glioma tissues.MethodsFresh tissues of 36 patients with glioma，including 21 cases of low malignance (grade Ⅰ~Ⅱ) and 15 cases of high malignance (grade Ⅲ~Ⅳ)，were used in the present study.The expression of EGFL7 mRNA in these glioma tissues was examined by RT-PCR. Other 45 cases of formalin fixed，paraffin embedded specimens of glioma，including low 24 cases of malignance (grade Ⅰ~Ⅱ) and 21 cases of high malignance (grade Ⅲ~Ⅳ)，were used to assess the expression status of EGFL7 protein by immunohistochemical staining. The correlation between EGFL7 expression and glioma grades was investigated.ResultsThe expression of EGFL7 mRNA and protein with human gliomas was detected. EGFL7 protein located in plasm of tumor cells and endothelial cells，and its expression correlated directly with the degree of malignancy(t＝4.399，P<0.01).However，it was not detected in normal brain.ConslusionTumor cells and endothelial cells in human glioma tissues can express EGFL7 gene，and the level of EGFL7 expression directly correlates with the degree of malignancy. It suggested that EGFL7 may contribute to the rapid growth and invasion of gliomas.
