骨髓间质干细胞诱导成骨细胞成骨能力的实验研究

摘    要:目的:研究人骨髓间质干细胞(BMSCs)诱导成骨细胞的成骨能力,为组织工程化骨应用于临床提供理论依据.方法:制备人BMSCs与犬脱细胞-脱钙骨的复合物,将复合物回植于裸鼠皮下作为实验组,同时植入空白材料作为对照组,分别于植入4、8、12周取材,进行大体观察和组织切片HE染色观察,分析其体内成骨能力.结果:大体观察:随着植入时间延长,实验组体积变化不明显,12周时材料与背部皮肤粘连;对照组体积略缩小,材料基本降解.组织切片HE染色观察见对照组无新骨形成,有纤维包裹,实验组8、12周时有新骨小梁形成,周围有较多小血管形成.结论:人BMSCs诱导的成骨细胞具有良好的体内成骨能力,脱细胞-脱钙骨松质-人BMSCs-成骨细胞是较理想的组织工程化骨.

Abstract:Objective: To study the human bone marrow mesenchymal stem-cells inducing the osteogenic activity of osteoblast and provide theoretical evidence for clinical use. Methods: Put the decell-decalcification spongy bone-human BMSCs-osteoblast compound into nude mice. Meanwhile transplant vacant material as the control group and took out to observe and histology HE staining after 4 weeks, 8 weeks, 12 weeks respectiveyly, and then analyzed their osteogenic activity in vitro. Results: Microscopic observed with the extend of implant time there were not volume change in experiment group, and material adhered to the back skin after 12 weeks. Volume slightly reduce in control group owing to material degeneration. There was not new bone formation but fabric packing in experiment group by observed histology slide HE staining. New bone trabecula formation could be observed in experiment group, after 8~12 weeks. There were little blood vessel on periphery. Conclusion: Human BMSCs after being induced have the strong osteogenic ability, and the compounds of decell-decalcification spongy bone-human BMSCs-osteoblast is a kind of ideal ossification of tissue engineering.
