氯通道阻断剂对H_2O_2诱导的胰岛RIN-mβ细胞凋亡的影响

摘    要:目的观察氯通道阻断剂对H2O2诱导的胰岛RIN-mβ细胞凋亡的影响,探讨氯通道在RIN-mβ细胞凋亡中的作用。方法采用H2O2诱导的胰岛RIN-mβ细胞凋亡模型,观察氯通道阻断剂(DIDS、NPPB和NFA)对细胞存活率、形态学变化、凋亡的影响。结果氯通道阻断剂单用时对胰岛RIN-mβ细胞活力无明显影响,但能明显提高H2O2处理的胰岛RIN-mβ细胞的存活率。与模型对照组相比,氯通道阻断剂DIDS、NPPB和NFA对抗组细胞存活率明显增加(P<0.01),细胞凋亡率明显降低(P<0.01)。结论氯通道阻断剂对H2O2诱导的RIN-mβ细胞损伤和凋亡具有明显的保护作用。

Abstract:Aim To investigate the effects of chloride channel blockers on the apoptosis of RIN-mβ cells of pancreatic islet induced by H2O2.Methods The apoptotic model was made by H2O2 exposed for six hours with a concentration of 500 μmol·L-1.The chloride channel blockers:DIDS,NPPB and NFA were administered to pretreat the samples respectively.The cell viability,morphological changes,and apoptosis rate were observed.Results Chloride channel blockers alone have no marked effects on the cell viability of RIN-mβ cell.However,they elevated the cell viability of RIN-mβcell disposed of by H2O2.Compared to H2O2 group,the groups of DIDS +H2O2,NPPB+ H2O2 and NFA+H2O2 have significant difference in cell viability and apoptosis rate(P<0.01).Conclusions Chloride channel blockers(DIDS,NPPB and NFA) remarkably inhibit apoptosis of RIN-mβ cells induced by H2O2.
