大鼠分流叠加炎症肺高压模型的建立

摘    要:目的建立大鼠分流叠加炎症所致肺动脉高压模型,探讨分流叠加炎症肺高压模型可行性。方法对分流组及分流叠加炎症组大鼠行腹主动脉-下腔静脉分流术,分流叠加炎症组于每周第3、6天予温水擦洗尾部并常规消毒,经尾静脉缓慢注射0.6%FeCl3(0.2mL/100g体重)。术后6周和11周,分别测定肺动脉平均压(PAMP)、右心室(RV)/左心室+室间隔(LV+S)比值,观察肺血管结构的变化。结果6周和11周分流叠加三氯化铁组大鼠肺动脉平均压、右心室肥厚度及肺血管平滑肌细胞增生程度均比分流组重,差别具有显著性(P〈0.05)。结论本模型更好反映肺动脉高压发病过程分流和炎症因素的相互作用。

Abstract:[Objective] To establish a rat model of pulmonary hypertension induced by left to right shunt and reflecting of inflammatory effects. [Methods] Abdominal aorta and inferior vena cava shunting was produced in rats, intravenous injection of ferric chloride (0.6%, 0.2 mL/100 g BW) 2 times 1 week were given to rats of the shunt and inflammation group. Pulmonary artery mean pressure(PAMP) of each rat was measured by using a right cardiac catheterization procedure. The radio of right ventricular mass to left ventricular plus septal mass (RV/LV+S) was detected. The structural changes in pulmonary arteries were observed. [Results] PAMP, RV/LV+S and PASMCH were significantly increased in rats affected by inflammation. [Conclusion] The model of pulmonary hypertension by shunt and intravenous injection of ferric chloride can be used to study the roles of inflammation playing in the pathogenesis of pulmonary hypertension.
