MDM2蛋白在喉鳞状细胞癌中的表达意义

目的　初步探讨 MDM2在喉癌发生发展中的机制 .方法　采用链霉菌抗生物素蛋白 -过氧化物酶 (S- P)免疫组织化学法 ,检测 1 2例声带息肉 ,1 5例喉癌旁组织 ,1 1例喉不典型性增生及 41例喉鳞状细胞癌组织中 MDM2蛋白的表达 ,并研究蛋白表达与肿瘤病理分级、临床分期、临床分型、淋巴结转移的关系 .MDM2蛋白阳性表达为核着色 .结果　 MDM2蛋白表达阳性率在各个组分别为声带息肉 0 .0 % ,喉癌旁组织40 .0 % ,喉不典型性增生 72 .7% ,喉鳞状细胞癌组织 39.0 % .MDM2蛋白表达在声带息肉组与癌旁组织组、声带息肉组与喉不典型性增生组、声带息肉组与喉鳞状细胞癌组织组、喉不典型性增生组与喉鳞状细胞癌组织组 (P<0 .0 5)均有显著性差异 .喉鳞状细胞癌组 MDM2蛋白表达与临床分期有关 ,与病理分级、临床分型、淋巴结转移无关 .结论　 MDM2基因在喉癌发生、发展中有重要作用 ,其异常表达是喉癌发生中早期分子事件 ,检测 MDM2蛋白有助于喉癌早期诊断

Expression and clinical significance of MDM2 protein in laryngeal squamous cell carcinoma

AIM To investigate the significance of biological behaviorion of MDM2 gene in laryngeal squamous cell carcinoma (LSCC). METHODS Expression of MDM2 protein levels were assessed by immunohistochemical method (S P) in 12 specimens of polyp of vocal cord (PVD), 15 of adjacent normal tissues (ANT), 11 of laryngeal atypical hyperplasia (LAD) and 41 of laryngeal squamous cell carcinoma (LSCC). RESULTS Percentage of detectable MDM2 expression was 0.0%, 40.0%, 72.7%, 39.0% respectively. The results showed MDM2 expression in PVD was significantly lower than that in ANT, LAD and LSCC (P). There was significantly different from LAD and LSCC. MDM2 expression in LSCC was correlated with clinical stage but not with pathological grade , clinical tape and lymph node metastases. CONCLUSION MDM2 gene plays an important role in occurrence and development of carcinogenesis of larynx so that MDM2 could be an early molecular event in the carcinogenesis of larynx. Detection of MDM2 protein would be helpful for the diagnosis of laryngeal carcinoma.
