Hedgehog通路中PTCH1蛋白在人胃癌组织中的表达及其临床意义

目的探讨Hedgehog通路中PTCH1蛋白在人胃癌组织中的表达及其与胃癌发生的关系。方法应用免疫组化法检测PTCH1蛋白在40例胃癌及癌旁组织中的表达情况以及分布规律,分析PTCH1蛋白的表达与胃癌的临床生物学特征的关系。结果 PTCH1蛋白在胃癌组织中的表达比癌旁组织低（50.0%vs87.5%）,差异有高度统计学意义（P〈0.01）。PTCH1蛋白表达与胃癌肿瘤大小和转移无相关性,与肿瘤分化程度有关,低分化癌组织中PTCH1蛋白表达率明显低于中高分化癌组织中表达率（41.4%vs72.7%）,差异有统计学意义（P〈0.05）。结论 PTCH1蛋白在胃癌组织中的低表达与胃癌的发生、发展密切相关,提示PTCH1蛋白检测有可能成为临床新的胃癌的早期诊断标志物并可预测患者的预后及转归。

Expression and Its Clinical Significance of PTCH1 Protein Involved in Hedgehog Signal Pathway in Human Gastric Carcinoma

Objective To observe the expression of PTCH1 proten in human gastric carcinoma and analyse the relationship between its expression and gastric carcinoma patients＇ clinical characteristics.Methods The expression differences of PTCH1 proten in 40 cases of human gastric carcinoma tissues、 adjacent normal tissues were detected by the methods of immunohistochemistry.Results Gastric carcinoma tissues and adjacent normal tissues expressed PTCH1 proten,the rate of expression of PTCH1 proten in gastric carcinoma tissues and adjacent normal tissues was 50.0%（20/40）、87.5%（35/40） respectively,and the rate of expression in the adjacent normal tissues was significantly higher than that in the gastric carcinoma tissues（P0.01）.The expression level of PTCH1 proten was significantly correlated with the differentiation degree of gastric carcinoma（P0.05））.The expression level of PTCH1 proten in well-moderate differentiation gastric carcinoma tissues and poor differentiation gastric carcinoma tissues was 72.7%（8/11）、41.4%（12/29） respectively.Conclusion The low expression of PTCH1 proten in gastric carcinoma may play a role in gastric cancer.It is possible that the detection of PTCH1 proten may be a novel method for the diagnosis of gastric carcinoma and a new clue to indicate the prognosis.
