星形胶质细胞机械性损伤后内皮性脂酶的表达

摘    要:目的研究星形胶质细胞划痕损伤后内皮性脂酶(EL)的表达情况。方法取新生大鼠脑皮质,体外分离培养、纯化星形胶质细胞,对其进行划痕损伤,取损伤后不同时间点0 h、1 h、3 h、6 h、12 h、24 h、48 h及正常对照,用Western blot和免疫组化标记检测EL的表达变化及其与凋亡促进因子caspase-3的关系。结果与正常对照组比较,胶质细胞在损伤后6 h EL的表达增高十分明显(P〈0.01),在损伤后1h出现caspase-3的表达增高(P〈0.01)。结论星形胶质细胞机械性损伤后1 h出现caspase-3表达增高,随后出现EL表达增高,提示星形胶质细胞于损伤后早期即出现凋亡现象,EL在中枢神经系统损伤后修复与再生过程中起重要作用。

Abstract:Objective To study the expression of endothelial lipase after the mechanical injury of rat primary astrocytes in vitro.Methods Purified cortical astrocytes were prepared from neonatal rats.The astrocyte mechanical injury model was established,the expression of endothelial lipase and the relation between the expression of caspase-3 in injured astrocytes at the 0 h,1st h,3rd h,6th h,12th h,24th h,48th h and the normal astrocytes were observed and examined by immunocytochemical staining and Western blot assay.Results Western blot showed that the expression of endothelial lipase increased 6 h after mechanical injury,caspase-3 increased 1 h after mechanical injury.Conclusion Caspase-3 increases 1 h after astrocytes mechanical injury,then the endothelial lipase expresses at a high level,which indicates that the apoptosis occurs in astrocytes at the early stage after mechanical injury,endothelial lipase may have important implications in the repair following injury and regeneration of central nervous systerm.
