生物素标记IL－6基因探针检测MOFE大鼠外周血IL－6mRNA表达水平

将光敏生物素标记含目的基因的质粒ＤＮＡ，用ＤＮＡａｓｅ随机切成１００－５００ｂｐ大小标记探针与待测老龄大鼠多衰模型中外周血的单核淋巴细胞在适当条件下进行杂交，应用原位核酸分子杂交方法，鉴定分析老龄大鼠多器官衰竭中ＩＬ－６ｍＲＮＡ表达水平。实验结果显示：静脉注射油酸造成急性肺损伤后，再注射小剂量内毒素可造成老龄大鼠ＭＯＦＥ模型；ＩＬ－６基因探针对于外周血有敏感的杂交信号，随着损伤加强ＩＬ－６ｍＲＮＡ表达递增。结果表明ＩＬ－６基因探针可用于检测多器官衰竭的程度。

The photobiotin-labelled probe of IL-6 gene for detecting the exprission level of IL-6mRNA in the peripheral lymphocyte of aged rats with multiple organ failune in the elderly

The photobiotin-labelled probe of IL-6 gene was used to detect in the expression level of IL-6mRNA in the peripheral lymphocyte of aged rats with MOFE in situ hybridization. Oleic acid was intravenously injected into aged rats to produce acute lung injury , Onthe bases of this acute lung injury model, E. coli endotoxin was subsequently injected to induce multiple organ failure. The exprements showed that the IL-6 gene probe nad sensitive hybridizable signal in peripheral lymphocytes, and the level of IL-6mRNA expression paralled with the degree of injury. The results suggest that the IL-6 gene probe may be used for the identification of the different period of MOFE.
