尿酸对人脐静脉内皮细胞增殖及PAI-1分泌的影响

摘    要:目的探讨尿酸(UA)对人脐静脉内皮细胞(HUVECs)增殖及纤溶酶原激活物抑制物-1(PAI-1)分泌的影响,以阐明其在动脉粥样硬化发生中的作用。方法用MTT比色法观察不同浓度UA(0、2、4、8、16mg/dL)作用24h对HUVECs增殖的影响;用ELISA方法评价不同浓度UA(0、2、4、8、16mg/dL)作用HUVECs 24h及8mg/da UA作用HUVECs 1、3、6、12、24h对细胞培养液中PAI-1的影响。结果UA呈剂量依赖性抑制内皮细胞增殖,8、16mg/dL UA组与对照组(0mg/dLUA)比较均有统计学意义(P〈0.01)。UA诱导HUVECs分泌PAI-1呈浓度依赖性增加,8、16mg/dL UA组与对照组比较均有统计学意义(P〈0.01);UA诱导HUVECs分泌PAI-1亦呈时间依赖性,与1h比较,3、6、12、24h均有统计学意义(P〈0.05或P〈0.01)。结论UA抑制HUVECs增殖,诱导HUVECs分泌PAI-1,这可能与高尿酸血症致动脉粥样硬化作用机制有关。

Abstract:Objective To investigate the effects of uric aeid(UA) on the production of plasminogen activator inhibitor-(PAI-1) and the proliferation of cultured human umbilical vein endothelial cells(HUVECs). Methods The effects of UA(0,2,4,8,16 mg/dL) on the proliferation of HUVECs was observed after 24 hours by MTT assay. HUVECs were incubated with different concentration (0,2,4,8,16 mg/dL)of UA for 24 h and 8 mg/dL of UA for different periods (1, 3,6,12,24 h). PAI-1 in supernatant medium was detected by cell ELISA. Results UA inhibited the proliferation of H UVECs in a dose-dependent manner, compared with control group(0 mg/dL UA),UA(8,16 rag/alL)group could inhibit the proliferation of HUVECs(P〈0.01). PAI-1 activated by UA in HUVECs remarkably increased in a dose-dependent manner,compared with control group,UA(8, 16 rag/alL)group could enhance the production of PAI-1 in HUVECs(P〈 0.0l). The enhancing effect was also time-dependent. Compared with 1 h, the levels of PAI-1 were significantly increased at 3,6, 12,24 h of stimulation with UA(P〈0. 05 or P〈0. 01). Conclusion UA could obviously inhibit the proliferation of HUVECs and stimulate the produe-tion of PAI-1 in HUVECs, which may thereby play an important role during the atherogenesis.
