凋亡相关基因Fas、FasL和bcl-2在膀胱移行细胞癌中的表达

摘    要:目的 研究凋亡相关基因Fas、FasL和bcl-2 ,在膀胱移行细胞癌 (TCC )中的表达情况 ,并探讨其临床意义。方法 采用S -P免疫组化方法 ,检测 67例TCC及 12例正常膀胱组织中Fas、FasL和bcl -2蛋白的表达。结果 在TCC和正常膀胱组织中FasL蛋白阳性率分别为 5 6.7%和 0 ,两者有非常显著性差异 (P <0 .0 1) ,而Fas和bcl -2蛋白在TCC和正常膀胱组织中的表达无显著性差异 (P >0 .0 5 )。Fas和bcl -2蛋白表达阳性率在TCC不同病理分级、分期有非常显著性差异 (P <0 .0 1)。FasL蛋白表达随着TCC分级、分期的增加 ,呈降低趋势 ,但无显著性差异 (P >0 .0 5 )。结论 Fas/FasL系统异常可能是TCC发生发展过程中免疫逃逸的重要因素 ;在TCC发生以及在低分化和浸润性TCC中 ,Fas/FasL基因介导的细胞凋亡可能被bcl -2基因的过度表达所抑制 ;Fas和bcl -2基因表达可作为监测TCC恶性程度的重要指标

Abstract:Objective To investigate the expression of apoptosiss correlative gene Fas,FasL and bcl-2 in transitional cell carcinoma of bladder(TCC) and its significance.Methods 67 patients and 12 normal bladder tissues were analysed by S-P immunohistochemistry technique to determine Fas,FasL and bcl-2 expression.Results The positive rates of FasL protein were 56.7% and 0 in the TCC and normal bladder tissues respectively.The difference was statistically significant(P<0.01).The expression of Fas and bcl-2 did not show correlation between the two groups(P>0.05).Fas protein and bcl-2 protein expression correlated with histological grade and pathological stage of TCC(P<0.01).However,FasL protein decreased with the increase of tumor grade and stage without a significant difference(P>0.05).Conclusion The results suggest that the abnormal expression of Fas/FasL might play an important role in immune evasion of TCC.Overexpression of bcl-2 in TCC might inhibit Fas/FasL induced cell death.The expression of Fas and bcl-2 might be regarded as a marker to determine the severity in TCC.
