子宫内膜癌组织中细胞凋亡及相关基因的表达

摘    要:目的探讨载脂蛋白-al(Fas)及B淋巴细胞瘤/白血病-2基因(Bc l-2)对子宫内膜腺癌中细胞凋亡的作用。方法用免疫组化ABC法检测40例子宫内膜癌及36例子宫内膜不典型增生中Fas及Bc l-2的表达,TUNAL法检测细胞凋亡的发生。结果子宫内膜腺癌与子宫内膜不典型增生组织相比,细胞凋亡率显著降低,并且Fas表达减弱,而Bc l-2表达增强,差异均有统计学意义(P<0.05)。相关分析显示:细胞凋亡率与Fas表达呈正相关(r=0.62,P<0.05),与Bc l-2表达呈负相关(r=-0.56,P<0.05)。结论子宫内膜腺癌和子宫内膜不典型增生组织均可发生凋亡,Fas及Bc l-2基因参与子宫内膜腺癌的发生和发展。

Abstract:Objective To investigate the effects of apoptosis and related gene Fas and Bcl-2 in the endometrial carcinoma and dysplasia.Methods Fas and Bcl-2 were detected by immunohistochemistry and apoptotic cells were stained in situ by terminal deoxynucleotidyl transferase mediated-dUTP nick end labeling(TUNAL) in 40 cases of endometrial carcinoma and 36 cases of dysplasias.Results Apoptotic index in dysplasias was significantly higher than that in endometrial carcinoma(P<0.05);the expression of Bcl-2 was increased and Fas was decreased in endometrial carcinoma(P<0.05).Fas expression was positively and Bcl-2 expression was negatively associated with the rate of apoptosis(r=0.62,P<0.05;r=-0.56,P<0.05 respectively).Conclusions Our study indicates that both in the endometrial carcinoma and dysplasia tissues apoptosis could occur,Fas and Bcl-2 take part in the occurrence and progression of endometrial carcinoma
