摘    要:目的观察牛嗜铬细胞移植于PD样大鼠脑内的存活及功能。方法用6-OHDA损毁大鼠一侧黑质细胞制成偏侧PD样大鼠模型。在右侧纹状体内植入约2万个牛嗜铬细胞悬液。记录阿朴吗啡诱发大鼠的旋转行为。实验结束后取大鼠脑做组织形态学检查。结果大鼠在移植后阿朴吗啡诱发的旋转行为得到明显纠正(P<0.001)。移植后12周荧光组化显示移植区有大量SPG阳性细胞,其形态类似在正常肾上腺髓质内的嗜铬细胞的形态。结论脑内移植牛嗜铬细胞可改善PD样大鼠药物诱发的旋转行为;提示牛嗜铬细胞在移植12周后仍能在大鼠脑纹状体内存活。

Abstract:Objective: To observe the viability and function of bovine chromaffin cells transplanted in a rat modelof Parkinson's disease. Methods Rats with unilateral 6-hydroxydopamine lesions of the rightdopaminergic neurons of substantia nigra received intrastiatal bovine chromaffin cells grafts. Apomorphine-induced rotation were monitered in automated rotometer bowls for 4O min at various timepoints aftergrafting. The brain was cut for morphological analysis at the end of the experiments. ResultsApomorphine-induced rotational behavior were significantly reduced after grafting (P<0.01). There werea large numbers of survival bovine chromaffin cells in the host striatum 3 months postgrafting. ConclusionsThe present study demonstrates that intrastriatl implantation of bovine chromaffin cells in rats with 6-OHDA lesions can reduce Apomorphine-induced rotational behavior and survive for 3 months withoutimmunosupperssion.
