胰腺癌中TFPI-2基因的表达及其意义

摘    要:目的 探讨人胰腺癌中TFPI-2基因表达及其意义.方法 采用RT-PCR及Westernblot技术,检测TFPI-2基因在50例胰腺癌、35例转移灶、8例正常胰腺组织中的表达,同时用Boy-den小室及裸鼠皮下种植瘤模型检测胰腺癌细胞系Panc-1体内、体外侵袭及转移能力.结果 胰腺癌组织及转移灶中TFPI-2 mRNA及相应蛋白表达水平均低于正常胰腺组织(P<0.05).TFPI-2的表达水平与性别、年龄、肿瘤大小及组织学分级无关,而与临床分期、尤其是有无远处转移及神经侵犯有关.Ⅲ、Ⅳ期胰腺癌组织中TFPI-2表达水平低于Ⅰ、Ⅱ期(P<0.05);无远处转移、无神经侵犯的胰腺癌组织中TFPI-2表达水平高于有远处转移、有神经侵犯的胰腺癌组织(P<0.05).转染基因胰腺癌细胞系Pane-1-TFPI-2体外侵袭能力下降,无明显肌层浸润及肝、肺转移现象.结论 TFPI-2基因表达的降低与胰腺癌侵袭和转移有关,可能参与胰腺癌的侵袭和转移调控.

Expression of TFPI-2 gene in human pancreatic carcinoma and its clinical significance
Abstract:Objective To investigate the expression of TFPV2 in human pancreatic carcinoma and its clinical significance. Methods RT-PCR and Western blot were used to detect the expression of TFPI-2 in 50 pancreatic carcinoma, 35 metastasis, 8 normal pancreatic tissue. Boyden chamber and subcutaneous neoplasm model was used to evaluate the effect of TFPI-2 on invasion and metastasis in vitro and in vivo. Results The expression of TFPI-2 mRNA and TFPI-2 protein in pancreatic carcino-ma and metastasis was lower than that of normal pancreatic tissue (P<0.05). The expression of TF-PI-2 had nothing to do with gender,age,size of the tumor and the grade in histology but was related to clinical stage,especially distant metastasis and invasion of nerves. The expression of TFPI-2 in pancre-atic carcinoma in the clinical stage of Ⅲ, Ⅳwas lower than that in the clinical stage of Ⅰ, Ⅱ (P<0. 05). The expression of TFPI-2 in pancreatic carcinoma with distant metastasis and nerve invasion was lower than that of with no distant metastasis and nerves invasion (P<0.05). Boyden chamber as-say showed that the invasion ability was lower in Panc-1-TFPI-2 group than that in Panc-l-V group and Panc-1-P group. In Panc-1-P and Panc-1-V group, the neoplasm invaded into muscular coat and metastasized to liver and lung tissue. But in Panc-1-TFPI-2 group,there was no infiltration of muscu-lar coat and metastasis in liver and lung. Conclusion The expression of TFPI-2 is relevant to invasion and metastasis of pancreatic carcinoma. TFPI-2 may play an important role in regulation of invasion and metastasis of pancreatic carcinoma.
