针刺对实验性大鼠前列腺组织中Ki-67和bFGF表达水平的影响

目的观察针刺对前列腺增生症(BPH)的影响,探讨其作用机制。方法将雄性SD大鼠随机分为正常组、模型组、药物组、针刺组和针药结合组,采用去势后皮下注射丙酸睾丸酮4 mg/kg的方法来复制大鼠前列腺增生的模型,同时给予相应治疗,共进行30 d。造模和治疗结束后,测量各组大鼠前列腺湿质量指数、体积,光镜和电镜下观察前列腺组织的形态学变化,用免疫组化法检测Ki-67和bFGF在各组大鼠前列腺组织中表达的差异。结果模型组大鼠前列腺湿质量指数、体积及组织中的Ki-67和bFGF的表达水平与假手术组比较明显升高,腺上皮细胞数目与假手术组比较明显增加(P均<0.05);药物组、针刺组及针药结合组大鼠的前列腺湿质量指数、体积及组织中的Ki-67和bFGF的表达水平与模型组比较明显降低(P均<0.05),腺上皮细胞数目与模型组比较明显减少(P<0.05)。3个治疗组大鼠的前列腺湿质量指数、体积和组织中的Ki-67和bFGF的表达水平比较无显著性差异(P均>0.05)。结论针刺能够明显降低前列腺增生大鼠的前列腺湿质量指数和体积,抑制Ki-67和bFGF在前列腺组织中的表达,从而达到抑制前列腺增生的作用。

Experimental study on influence of acupuncture on Ki-67 and bFGF in prostatic tissue of rats with BPH

Objective It is to observe the influence of acupuncture on benign prostatic hyperplasia(BPH) and approach its mechanism.Methods Male rats were randomly divided into control group,model group,medicine group,acupuncture group and acu-medicine group.The BPH model was made by castrated and injected with testosterone propionate 4 mg/kg subcutaneously.At the same time,the rats were treated with respective methods for 30 days.Then the prostate damp weight index,prostate gland volume,Ki-67,bFGF,and the pathological changes of prostate gland in all groups were measured.Results Compered with control group,the prostate damp weight index,prostate gland volume,the levels of Ki-67 and bFGF,and the number of prostate epithelial cell of model group were significantly increased(P<0.05);while compared with model group,the prostate damp weight index,prostate gland volume,the levels of Ki-67 and bFGF,and the number of prostate epithelial cell of treated groups were significantly reduced(P<0.05).There was no significant difference between these three treating groups in the prostate damp weight index,prostate gland volume,and the levels of Ki-67and bFGF(P>0.05).Conclusion Acupuncture can significantly lower the prostate damp weight index and prostate gland volume,restrain the expression of Ki-67,bFGF in prostate hyperplasia rats,in order to restrain prostate hyperplasia.
