荧光定量PCR检测HBV-DNA与血清标志物的关系探讨

目的:探讨乙型肝炎血清HBV-DNA水平与乙肝抗原抗体模式的关系。方法:对320例血清标本同时用荧光定量PCR(FQ-PCR)检测HBV DNA和用酶联免疫吸附法(ELISA)检测乙肝标志物,根据不同检测结果进行对比分析。结果:320份血清中,乙型肝炎病毒标志物不同组合HBV-DNA阳性率有差异(P<0.01),320份血清中,有188例HBV-DNA阳性,占56.2%,HBsAg HBeAg HBcAb阳性者HBV-DNA阳性率占98.6%,>10copies/ml占64.5%,HBsAg HBeAb HBcAb阳性者HBV-DNA阳性率占37.5%,10 copies/ml占18.2%。结论:HBeAg和HBV-DNA有明显的相关性,PCR定量检测HBV-DNA含量更有助于判断体内HBV复制的情况及传染性强弱,在临床上有重要意义。

Compare the Relationship Between Hepatitis B Virus DNA Detected by Quantitative PCR and the HBV Serum Markers

Objective To study the relation between the quantity of HBV-DNA and hepatitis B virus markers in hepatitis B patients.Methods Hepatitis B virus DNA was detected by FQ-PCR and hepatitis B virus markers were detected by ELISA in 320 serum sample.Results The amount of HBV-DNA was different in 3 HBV immunological Markers groups of 320 patients with positive HBVM . 188 had positive serum HBV-DNA (positive rate 56.2%) The positive rate of HBV-DNA in patients with positive serum HBsAg,HBeAg and HBcAb was 98.6%,and the positive rate of HBV-DNA content greater than 10copies/ml was 64.5%. The positive rate of HBV-DNA in patients with positive HBsAg HBeAb and HBcAb was 37.5%and the the positive rate of HBV-DNA content greater than 10 copies/ml was 18.2%.Conclusion There is a significant correlation between HBeAg and HBV-DNA .The serum HBV-DNA level quantified by PCR is more helpful for detecting the replication and communicability of virus in hepatitis B patients.
