摘    要:目的探讨全反式维甲酸对体外培养子宫肌瘤细胞中COLⅠ、FN表达的影响以及对细胞增殖和凋亡的作用,为子宫肌瘤药物治疗提供新的实验依据。方法体外培养子宫肌瘤细胞,不同浓度全反式维甲酸作用后,应用MTT法检测细胞增殖活性,半定量RT-PCR检测COLⅠ、FN水平变化,流式细胞仪检测细胞凋亡率。结果低浓度(10-8mol/L)全反式维甲酸可降调肌瘤细胞中COLⅠ、FN表达,抑制肌瘤细胞的增殖及促进肌瘤细胞的凋亡(P〈0.05);且随着药物浓度的递增,COLⅠ、FN表达逐渐降低(P〈0.05),抑制肌瘤细胞增殖活性及促进肌瘤细胞凋亡率逐渐增强。结论全反式维甲酸以浓度依赖性方式,从核酸水平检测降调肌瘤细胞中COLⅠ、FN的表达,抑制体外培养肌瘤细胞的增殖且促进其凋亡。

Abstract:Objective To investigate the expression of collagenⅠ,fibronectin and all-trans retinoic acid on the proliferation and apoptosis of uterine leiomyoma cells in vitro.Methods Uterine leiomyoma cells were cultured in vitro,after the action of all-trans retinoic acid with different concentration,MTT was used to examine the proliferative activity of uterine leiomyoma cells,RT-PCR was used to determine the expression of the collagenⅠ,fibronectin mRNA,and the rate of apoptosis were determined by flow cytometry.Results Low dosage(10-8mol/L) of all-trans retinoic acid had effect on the expression of collagenⅠ,fibronectin mRNA(P0.05).and to inhibit the proliferation activity of uterine leiomyoma cells and promote the apoptosis of uterine leiomyoma cells(P0.05).The inhibition collagenⅠ,fibronectin mRNA were shown to decline gradually along with the increase in the concentrations of all-trans retinoic acid(P0.05).Furthermore,the inhibition were shown to decline and promotion were shown to incline gradually along with the increase in the concentrations of all-trans retinoic acid.Conclusion The all-trans retinoic acid could suppress the proliferation and promote the apoptosis of uterine leiomyoma cells in the dosage-dependent manner by down-regulating the expression of the collagenⅠand fibronectin,suggesting that the drug may possibly be effective in the clinical treatment of uterine leiomyoma.
