平调饮对小鼠H_(22)肝癌模型凋亡基因的影响

摘    要:目的:研究平调饮对H22荷瘤小鼠Bcl-2、P53和Bax凋亡基因的影响,从而探讨平调饮治疗原发性肝癌的作用机制。方法:昆明种雄性小鼠80只,接种H22肝癌细胞后,随机分为生理盐水组、中药组、西药组及中西医结合四组,每组20只。造模第2d灌胃给药,连续7d,第8d脱颈处死。将肿瘤取出制作小鼠肿瘤细胞切片,进行HE染色,观察病理变化;用免疫组织化学方法对肿瘤中的Bcl-2、P53和Bax蛋白进行染色。用SPSS12.0软件统计结果。结果:造模后西药组小鼠状态较差,其体重明显低于中药组及荷瘤组(P0.01)。HE染色中药组的肿瘤细胞体积缩小,核固缩,核分裂相明显少于模型组,肿瘤组织可见片状坏死区。中药组和中西医结合组的Bcl-2和P53蛋白表达率明显降低,Bax蛋白的表达率则明显增高,与对照组比较有显著性差异(P0.01)。结论:平调饮对肝癌细胞具有抑制作用,其机制可能为通过对肿瘤细胞内P53、Bcl-2、Bax蛋白表达的改变,使肝癌细胞发生细胞凋亡,从而抑制肿瘤生长。

Abstract:Objective:To investigate the anti-tumor effect of Pingtiaoyin to apoptotic gene bcl -2、P 53 and bax in mice H22 and its mechanism on programmed cell death (PCD),and discuss the related mechanism to primary hepatic carcinoma (PHC). Methods:After 80 male KM mice were injected H22 tumor cells,they were divided into 4 groups:the model group(A),the chinese medicine group(B),the western medicine group(C),the combining traditional Chinese and western medicine group ( D). From the next day after injected,the groups were repectively fed by medicine,once daily for 7 days. The mice were killed in the eighth day and tumors were excavated and weighted. The H22 tumor from mice was collected and the anti-tumor rate calculated; make the slide of the H22 tumor from the mice and to observe by HE technique;noted the pathological changes;the expression of Bax ,Bcl -2 and P 53 protein in the tumor tissue was studied by immuno histo-chemical method and studied the result by spss12. 0. RESULTS:After the animal experiment,the weight of the mice in C group was lighter than that of the other groups(P 0. 05). The tumor cells of group B smaller in size,karyopyknosis is significantly less than the model set,tumor tissue was found sheets of necrotic area. The expression frequency of Bcl -2 and P 53 protein of B、D group were lower than A group(P 0. 05),that of Bax protein of B、D group were significant higher than it,significant difference was found between these groups (P 0. 05). CONCLUSION:Pingtiaoyin can inhibit the Hepatoma cell,its mechanism may be induced apop-tosis of Hepatoma cell by changing the expression of Bax ,Bcl -2 and P 53 protein in the tumor tissue ,there-by inhibiting tumor growth.
