摘    要:目的 观察苦参碱对人肝癌细胞株HepG2体外增殖和端粒酶活性的影响,进一步探讨苦参碱的可能作用机制.方法 采用人肝癌细胞株HepG2作为研究对象,MTT法检测不同浓度和作用时间的药物对细胞增殖的抑制作用;流式细胞仪分析细胞周期的分布及凋亡;PCR-ELISA法检测端粒酶活性的变化.结果 苦参碱具有抗HepG2细胞增殖、阻滞细胞周期、诱导凋亡及抑制端粒酶活性的作用.结论 对端粒酶活性的抑制可能是苦参碱发挥抗肿瘤作用的机制之一.

Abstract:Objective To investigate the anti-cancer mechanism of matrine by observing their effects on proliferation and telomerase activity. Methods The inhibition of proliferation in HepG2 cells were estimated by MTT-assay. Distribution of cells cycle and apoptosis were analysed using flow cytometry. The changes of telomerase activity were detected by PCR-ELISA method. Results Matrine could inhibit the proliferation of HepG2 cells, block cell cycle,induce apoptosis and inhibit the activity of telomerase. Conclusion The inhibition of telomerase activity may be one of the mechanisms of the anti-cancer effect of matrine.
