摘    要:目的:探索建立流式细胞术(FCM)检测哌拉西林/他唑巴坦抗铜绿假单胞菌效应的方法。方法:以碘化丙啶(PI)为荧光染料,用FCM检测铜绿假单胞菌标准菌株ATCC27853在不同浓度哌拉西林/他唑巴坦(TZPP)作用下各管平均荧光强度(MFI),并寻找其最佳试验条件。结果:TZPP浓度范围为0·125~64μg·mL-1时,细菌MFI值随药物浓度的升高而呈上升趋势。结论:FCM可用于检测药物抗铜绿假单胞菌的效应,并为进一步的药敏试验提供基础。

Abstract:OBJECTIVE: To establish a flow cytometry (FCM) method for the detection of anti-Pseudomonas aeruginosa (PA) effect of Piperacilline/Tazobactam. METHODS: With propidium iodide (PI) taken as fluorchrome, the mean fluorescence intensity (MFI) of Pseudomonas aeruginosa standard strain ATCC 27853 in different concentrations of Piperacilline/Tazobactam were detected by FCM, and the experiment condition was optimized. RESULTS: At a concentration range of 0.125~64μg·mL-1 for TZPP, MFI value of bacteria showed an increase tendency with the increase of blood drug concentration. CONCLUSION: FCM can be used to detect the anti-PA effect of drugs and provide the basis for further drug susceptibility test.
