摘    要: 目的研究神经生长因子(Nerve growthfactor,NGF)对人血管内皮细胞的细胞周期和对增殖细胞核抗原(Prolifer-ating cell nuclear antigen,PCNA)表达的影响.方法将NGF作用于体外培养的HUEVC 304细胞,利用流式细胞仪检测其细胞周期、DNA相对含量的变化,利用免疫组织化学的方法检测对PCNA表达的改变.结果NGF作用组出现C0~G1期细胞数量明显减少,S期细胞、G2~M期细胞明显增加,DNA相对含量无明显变化,PCNA表达明显增加(P<0.05).结论NGF能明显促进HEVC增殖.

Abstract:Abstract Objective To investigate the effect of nerve growth factor(NGF) on human vascular endothelial cell cycle and expression of proliferating cell nuclear antigen(PCNA). Methods NGF effected human umbilical vein vascular endothelial cells in the third passage were used. The cell cycle and DNA content of the endothelial cells were examined by flow cytometry. The expression of PCNA was assessed using im-munohistochemical method. Results Compared with the control groups, the cell cycle of NGF effected groups showed significantly increased percentages of Gj - M phase and S phase cells, and significantly decreased percentages of G0 - G1 phase cells ( P < 0.01). The contents of DNA were constant. The expression of PCNA was stronger than that in the controls ( P < 0.05). Conclusions NGF significantly accelerates proliferation of human vascular endthelial cells.
