咖啡因在骨肉瘤顺铂化疗中增效作用的实验研究

摘    要:目的探讨咖啡因在骨肉瘤细胞株(OS-732)顺铂化疗中的增效作用.方法骨肉瘤细胞株分别经咖啡因、顺铂、咖啡因 顺铂作用72h,采用MTT法体外细胞毒性试验观察三者的细胞毒性;应用流式细胞仪分析咖啡因、顺铂及两者配伍对骨肉瘤细胞株细胞周期的影响.结果骨肉瘤细胞株与浓度分别为0.2、2.0、5.0、10.0、20.0mmol/L的咖啡因作用时,细胞毒性指数(CI)分别为12.50%、27.88%、30.77%、56.73%、84.62%;与浓度为0.1、1.0、10.0、100.0μg/ml的顺铂作用时,CI分别为24.04%、54.80%、65.38%、88.46%;而与低浓度咖啡因与顺铂联合作用,其CI明显增加,随着各自浓度的升高,CI明显增加.流式细胞仪分析显示咖啡因对细胞周期也有明显的影响,使G0与G1期细胞比例明显增加,G2与M期细胞比例明显减少,S期细胞比例无明显改变.结论咖啡因在骨肉瘤细胞株顺铂化疗中有显著的增效作用,并能改变骨肉瘤细胞的细胞周期.

Abstract:Objective To observe the enhancing effect of caffeine on cisplatin in treatment of osteosarcoma cell line OS-732. Methods The osteosarcoma cell line OS-732 was incubated with different concentrations of caffeine,cisplatin and caffeine+cisplatin for 72h.With 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay the cytotoxicity was observed;and the cell cycle phases were analyzed by flow cytometry (FCM). Results The cytotoxicity index (CI) increased as the concentrations of caffeine or cisplatin increased.When the cells were treated with the combination of caffeine and cisplatin,the CI increased more prominently.FCM showed a marked increase of G0 / G1 fraction and decrease of G2 / M fraction of the osteosarcoma cells treated with caffeine. Conclusion Caffeine combined with cisplatin have an enhancing cytotoxicity on osteosarcoma cell line and also can change the cell cycle of the treated osteosarcoma cells.
