猴免疫缺陷病毒急性及慢性感染淋巴结的病理变化

摘    要:【目的】观察猴免疫缺陷病毒(SIV)急性及慢性感染猴淋巴结的病理改变,以掌握评价抗艾滋病药物、疫苗及免疫调节剂疗效指标,并探索猴艾滋病毒感染及猴艾滋病(SIV/SAIDS)的发病机制。【方法】采用SIVmax病毒急性及慢性感染恒河猴40只,先后分别在不同时间作淋巴结活检68例次及尸检死亡猴7只的淋巴结,对淋巴结进行病理常规切片、染色、光镜检查。【结果】淋巴结的病理变化可分为3个型:(1)增生型(感染后3~6或9个月);(2)退化型(感染后6~12或14个月);(3)耗竭型(11个月以上及部分中途死亡猴)。猴SIV感染的淋巴结病变与人HIV/AIDS的淋巴结的病变比较,两者的病变极为相似。【结论】猴SIV感染淋巴结病变的3个型可以作为猴艾滋病发生、发展和恶化的观察指标,SIV感染猴模型可以作为抗艾滋病药物、疫苗和免疫调节剂的疗效评价的动物模型和研究艾滋病发病机制的理想动物模型。

Abstract:Objective] Pathological changes of lymph nodes in rhesus monkeys with acute and chronic infection of simian immunodeficiency virus (SIV) were observed to evaluate the therapeutic effect of anti-AIDS drugs, vaccine and im-munoregulators and to explore the pathogenesis of SIV infection and simian AIDS. Methods] Biopsy of lymph nodes from 40 rhesus monkeys with acute and chronic infection of SIVmac were operated for 68 times in different time and postmortem examination of lymph nodes were also carried out in other 7 monkeys. The pathologic slide specimens were prepared by routine method. Results ] Pathological changes of lymph nodes under light microscope can be classified into 3 types: (l) proliferation (3 - 6 or 9 months after infection); (2) degeneration (6 - 12 or 14 months after infection); (3) failure (over 11 months after infection and in monkeys died of simian AIDS). A general idea of the progress of SIV infection in the monkeys were obtained by the examination of the lymph nodes. Similar pathological changes of lymph nodes were found in SPV-infected rhesus monkeys as compared with those in HIV/AIDS patients. Conclusion] The three types of pathological changes of impaired lymph nodes can be used as the criteria of occurrence, development and deterioration of simian AIDS respectively. SIV monkey models can be applied to evaluate the therapeutic effect of anti-AIDS drugs, vaccine and immunoregulators and to explore the pathogenesis of SIV infection and simian AIDS.
