不同浓度奥沙利铂对胃癌细胞中p53蛋白表达及胃癌细胞增殖的影响

摘    要:目的探讨不同浓度奥沙利铂对胃癌细胞中p53蛋白表达情况及胃癌细胞增殖的影响。方法选取人胃腺癌细胞株BGC-823,采用不同浓度的奥沙利铂处理BGC-823细胞后,采用CCK8法检测细胞的增殖抑制率。观察p53蛋白在胃癌细胞中的表达情况。结果奥沙利铂处理24、48、72 h时,随着奥沙利铂作用浓度(3.2、6.4、12.8、25.6μg/ml)的增高,胃癌细胞的增殖抑制率均逐渐增高,且同一浓度下的奥沙利铂处理时,随着作用时间的延长,胃癌细胞的增殖抑制率均逐渐增高。奥沙利铂处理24、48、72 h时,随着奥沙利铂作用浓度(3.2、6.4、12.8、25.6μg/ml)的增高,胃癌细胞中p53蛋白的表达水平逐渐增高,且同一浓度下的奥沙利铂处理时,随着作用时间的延长,胃癌细胞中p53蛋白的表达水平逐渐增高(P﹤0.05)。结论不同浓度奥沙利铂对胃癌细胞中p53蛋白的表达及胃癌细胞的增殖抑制率均具有影响。

Abstract:Objective To investigate concentration-dependent effects of oxaliplatin on the expression of p53 protein and proliferation of gastric cancer cell.Method The human gastric adenocarcinoma cell line BGC-823 was subjected to oxaliplatin treatment at different concentrations,the proliferation inhibition rate was determined by CCK8 assay,and the expression of p53 in gastric cancer cells was observed.Result After treatment by oxalipatin for 24,48,and 72 h,higher proliferation inhibition rate in gastric cancer cells was observed as the concentration of oxaliplatin increased(3.2,6.4,12.8,and 25.6μg/ml),additionally,similar trend was noted in each specific concentration with prolonged treatment.Within the same treatment time frame and increase of concentrations,the expression of p53 protein in gastric cancer cells increased in a oxalipatin concentration-dependent manner,and was also promoted as treatment time extended(P<0.05).Conclusion Oxaliplatin affects the expression of p53 protein and the cell proliferation in gastric cancer cell in a concentration-dependent manner.
