牙龈癌微血管密度及血管内皮生长因子表达的意义

摘    要:目的 :研究牙龈鳞状细胞癌组织中微血管密度 (MVD)及血管内皮生长因子 (VGEF)的表达和意义。方法 :采用CD3 4单克隆抗体和VEGF多克隆抗体 ,用免疫组化S -P法对 42例牙龈鳞状细胞癌标本进行免疫组化染色。结果 :牙龈癌组织MVD与正常牙龈组织MVD两者差异有显著性 (P <0 .0 0 5)。有淋巴结转移的牙龈癌MVD与无淋巴结转移的牙龈癌MVD ,两者之间差异有显著性 (P <0 .0 5)。牙龈癌组织中VEGF表达阳性率为 71.43 % ,正常牙龈组织中VEGF表达阳性率为 2 5.0 0 % ,两者之间差异有显著性 (P <0 .0 5)。有淋巴结转移组VEGF阳性率为 83 .3 3 % ,高于无淋巴结转移组VEGF阳性率 (66.67% ) ,两者之间差异无显著性 (P >0 .0 5)。VEGF阳性牙龈癌MVD高于VEGF阴性牙龈癌MVD ,两者之间差异有显著性(P <0 .0 2 )。结论 :肿瘤的血管生成在牙龈癌的发生过程中起一定作用。MVD与牙龈癌淋巴结转移及VEGF表达有密切联系

Microvessel density and expression of vascular endothelial growth factor in squamous cell carcinomas of gingiva

Abstract:Objective:To determine the effects of angiogenesis in c ervical lymph node metastasis and progression of squamous cell carcinomas of gin giva (GSCC), and the distribution and expression of vascular endothelial growth factor (VEGF) in GSCC.Methods: Paraffin sections of surg ically obtained GSCC samples from 42 patients were stained with CD34 monoclon al antibody by immunohistochemical S P method to demonstrate blood vessels. Exp ression of VEGF was tested with the S P method, and the microvessel density (M VD) was determined according to the percent of the neoplastic cells showing VEG F immunoreactivity and the degree of staining.Results: The MVD in GSCC was significantly higher than that in normal gingival tissue( P <0.005), The MVD of gingival SCC with lymph node metastasis was significantly h igher than that without nodal involvement ( P < 0.05 ). Positive expression of VEGF in GSCC was significantly higher than that in normal gingival tissue(7 1.43% vs 25.00 %, P <0.05). VEGF positive expression in GSCC with lymph n ode metastasis was not significantly higher than that without nodal involvemen t(83.33% vs 66.67%, P >0.05). MVD in VEGF positive samples of GSCC was sig nificantly higher than that in VEGF negative ones ( P <0.02).Conclu sions: There is a close association between angiogenesis and tumor p rogression in GSCC.MVD is correlated with lymph node metastasis and the expressi on of VEGF.
