精氨酸在大鼠动脉粥样硬化预防及治疗中的作用

摘    要:目的:通过精氨酸干预动脉粥样硬化大鼠来研究精氨酸对于大鼠动脉粥样硬化形成的预防和治疗作用。方法:将60只SD大鼠分成4组:对照组(10只)、高脂饮食组(30只)、辛伐他汀组(10只)及精氨酸预防组(10只)分别干预12周,每组各取10只处死;高脂组剩余20只随机分为模型组(10只)和精氨酸治疗组(10只),干预4周处死;检测血脂、一氧化氮合酶(NOS)、主动脉和肝脏病理。结果:各组大鼠血脂无明显差异;精氨酸预防组较高脂组总一氧化氮合酶、诱导型-氧化氮合酶下降(P<0.05);精氨酸预防组较高脂组大鼠主动脉内膜、中膜及内膜/中膜厚度均减小,中膜脂质沉积减轻(P<0.05),而精氨酸治疗组较模型组上述指标无明显差异;肝脏病理显示精氨酸预防组脂肪变性较高脂组减轻,而且肝细胞炎性改变较辛伐他汀组减轻。结论:口服补充精氨酸对减轻大鼠主动脉粥样硬化发生发展具有预防作用,但无明显治疗作用,主要是通过NO途径来实现的;精氨酸还可以减轻高脂饮食大鼠肝脏脂肪变性的程度,对肝脏损伤较辛伐他汀小。

Abstract:Objective: To investigate the effects of arginine on prevention and treatment of atherosclerosis in rats.Methods: A total of 60 Sprague-Dawley rats were assigned randomly to four groups as control group(n=10),high-fat diet group(n=30),Simvastatin group(n=10),and arginine prevention group(n=10),according to different treatment for 12 weeks.Twenty rats in high-fat diet group were assigned randomly to model group(n=10) and arginine treatment group(n=10) and fed for 4 weeks before sacrifice.Serum levels of lipids and NOS were detected,and aortal and hepatic changes were observed.Results: No significant difference was found in serum lipid levels among the six groups.The levels of total NOS and iNOS and the intima-media thickness and intima/media thickness ratio significantly decreased in arginine prevention group as compared with high-fat diet group(P<0.05),but those indexes were not different between model group and arginine treatment group.The hepatic pathology showed steatosis was alleviated in arginine prevention group as compared with high-fat diet group,while hepatic cells inflammatory changes was lessened in arginine prevention group rats as compared with Simvastatin group.Conclusion: The addition of arginine has significantly prevention effect on rat’s atherosclerosis development through nitric oxide pathway,while no therapeutic effect has found.A diet with supplemental arginine may reduce hepatic steatosis and even lessen hepatic damage as compared with Simvastatin.
