Claudin-4与MMP-2在大肠癌组织中的表达及其临床意义

目的:探讨紧密连接蛋白-4(Claudin-4)和基质金属蛋白酶-2(MMP-2)在大肠癌组织中的表达及其与肿瘤进展的关系。方法:应用免疫组织化学方法检测经病理证实的41例大肠癌、20例大肠腺瘤样息肉、16例正常大肠黏膜组织中Claudin-4和MMP-2的表达。结果:大肠癌、大肠腺瘤样息肉中Claudin-4、MMP-2阳性表达率显著高于正常大肠黏膜组织(P<0.05)。Claudin-4、MMP-2阳性表达率与大肠癌的浸润深度、淋巴结转移及Duke’s分期有关(P<0.05)。Claudin-4、MMP-2在大肠癌中的表达有相关性(P<0.05)。结论:Claudin-4和MMP-2的高表达可能促进大肠癌的发生,癌组织中Claudin-4的表达缺失和MMP-2过表达可能促进肿瘤的浸润和转移,两者的表达水平可能成为大肠癌早期诊断和预后的重要参考指标。

Expression of Claudin-4 and Metalloproteinase-2 in Colorectal Carcinoma and Their Clinical Significance

Objective: To explore the expression of Claudin-4 and matrix metalloproteinase-2(MMP-2) in colorectal carcinoma and their relationship with cancer progression.Methods: The expression of Claudin-4 and MMP-2 in 41 cases of colorectal carcinoma,20 cases of colorectal adenomatous polyp and 16 cases of normal colorectal mucosa were determined by SP technique of immunohistochemical staining with antibodies against Claudin-4 and MMP-2 respectively.Results: The positive rates of Claudin-4 and MMP-2 expression in colorectal adenomatous polyp and colorectal carcinoma were higher than those in normal mucosa respectively(both P<0.05).The positive rates of Claudin-4 and MMP-2 expression were related with infiltrating depth,lymph node metastasis and Duke’s stages of colorectal carcinoma(all P<0.05).The expression of Claudin-4 was positively correlated with the expression of MMP-2 in colorectal carcinoma(P<0.05).Conclusion: Overexpression of Claudin-4 and MMP-2 may be involved in the carcinogenesis of colorectal carcinoma,negative expression of Claudin-4 and overexpression of MMP-2 may promote the invasion and metastasis of carcinoma,and they are valuable markers in early diagnosis and prognosis of colorectal carcinoma.
