糖尿病小鼠玻璃体视网膜中Opticin的表达

摘    要:目的:观察opticin在1型、2型糖尿病小鼠及正常小鼠玻璃体视网膜mRNA和蛋白表达的变化。方法:50只雄性小鼠随机分为柠檬酸钠缓冲液对照组(A)10只,链脲佐菌素诱导1型糖尿病组(B)10只,2型糖尿病组30只,并将其随机分为1,2,3月组(C、D、E),各10只。于1,2,3月后处死2型糖尿病组小鼠,3月后处死1型糖尿病组小鼠和柠檬酸钠缓冲液对照组,取玻璃体视网膜组织,保存于液氮中。实时聚合酶链反应检测玻璃体视网膜中opticin mRNA表达,Western-blot检测opticin蛋白表达。结果:1型糖尿病组玻璃体视网膜的opticinmRNA表达量是正常组玻璃体视网的68%,P<0.01;2型糖尿病1,2,3月组玻璃体视网膜的opticin mRNA表达量分别是正常组玻璃体视网的37%,36%,27%,P<0.01;但1,2,3月组之间的差异无统计学意义,P>0.05;1型、2型糖尿病组玻璃体视网膜的opticin蛋白表达量较正常小鼠玻璃体视网膜显著下降,P<0.01。2型糖尿病的2月、3月组小鼠玻璃体视网膜mRNA和蛋白表达均低于1月组小鼠。结论:1型、2型糖尿病小鼠玻璃体视网膜中opticin的mRNA和蛋白表达量较正常小鼠显著减少,2型糖尿病组小鼠玻璃体视网膜mRNA和opticin蛋白表达随时间延长逐渐降低。说明opticin在糖尿病视网膜病变的发生机制中可能起着重要作用。

Abstract:Objective: To observe and compare the mRNA and protein levels of opticin in vitreous body and retina of normal and diabetic mice.Methods: Fifty male KM mice were randomly divided into five groups(10 mice in each group): control group(group A),streptozocin-induced type 1 diabetes mellitus group(group B),and type 2 diabetes mellitus groups for 1,2,and 3 month(s)(group C,D,E).The vitreous bodies and retina of the mice from each group were removed after mice were sacrificed for real-time PCR and Western blot assays.Results: The mRNA level of opticin in the vitreous body and retina of type1 diabetes mellitus mice was 68% of the normal control’s(P<0.01).The expression of opticin mRNA in vitreous body and retina of type 2 diabetes mellitus mice was 37%,36%,and 27% of the normal control’s(P<0.01).Expression levels of opticin protein in the vitreous body and retina of the type 1 and type 2 diabetes mice were also lower than in control ones.Conclusion: The content of opticin in vitreous body and retina of diabetes rats was significantly decreased as compared with the normal control ones,suggesting opticin might play a role in the development of diabetic retinopathy.
