氟他胺尿道下裂模型小鼠的睾丸形态与功能变化

摘    要:目的:采用雄激素受体拮抗剂氟他胺建立小鼠尿道下裂的动物模型,为进一步研究尿道下裂时小鼠睾丸组织形态及生精功能的变化提供理论及实验依据。方法:购7-8周龄BALB/c小鼠60只,体重20g,分成A、B、C、D 4组,每组10只。在母鼠孕第12-21天连续10d每天依次经皮下注射给予氟他胺0,30,60,90mg/kg。出生后5周观察有无尿道下裂与隐睾。测量肛门生殖器距离(AGD),并从每组取3只尿道下裂的雄性仔鼠解剖观察睾丸位置情况。并观察睾丸细胞形态变化及检测生精功能情况。结果:A-D组F1代小鼠尿道下裂发生率分别为0%,0%,30.77%,60.41%;隐睾发生率都为0%。睾丸组织细胞形态无明显变化,生精功能正常。结论:60mg/kg与90mg/kg的氟他胺能够诱导出F1代小鼠尿道下裂的动物模型,其睾丸细胞形态及生精功能变化不明显。

Abstract:Objective: To establish a mice model of hypospadias induced by flutamide,and to study the changes of testicular morphology and function.Methods: BALB/c mice with 7-8 weeks old and 20 g weight were divided into A,B,C,and D groups randomly.The mice in the four groups were consecutively injected with 0,30,60,or 90 mg/kg flutamide respectively subcutaneously per day for 10 days during gestation days 12 to 21.The anogential distance(AGD) was measured.The fetuses(F1 mice) from each group were anatomized to observe the position of testes and occurrence of hypospadias and cryptorchidism five weeks after birth.Furthermore,the morphological observation of the testicular cells and spermatogenesis testing were performed.Results: The incidence of F1 hypospadias in group A,B,C,and D was 0%,0%,30.77%,and 60.41% respectively,and none cryptorchidism was seen in all mice.Morphology of testicular cells and spermatogenesis changed mildly.Conclusion: Flutamide of 60 mg/kg and 90 mg/kg can induce hypospadias in F1 mice but the testicular structure and function is normal.
