缺氧诱导因子2α在妊娠高血压综合征胎盘中的表达及其意义

目的　通过观察缺氧诱导因子2α(HIF -2α)在妊娠高血压综合征和正常妊娠胎盘组织中表达的差异,探讨HIF -2α与妊娠高血压综合征发病过程的关系,为妊娠高血压综合征的诊治提供新的理论依据。方法　收集10例妊娠高血压综合征和10名年龄配对的正常妊娠胎盘组织,运用蛋白质印迹法(westernblot) ,检测HIF 2α蛋白的表达水平;应用实时聚合酶链反应(real timePCR)方法,检测HIF -2αmRNA的表达水平。结果　与正常妊娠胎盘组织相比,妊娠高血压综合征胎盘中HIF -2α蛋白和mRNA表达水平均增高。结论　HIF -2α在妊娠高血压综合征胎盘组织中的异常高表达,进一步证明缺氧在妊娠高血压综合征发病中的作用,其高表达可能与妊娠高血压综合征的病因及病理过程有关。

Expression of hypoxia-inducible factor 2α in placentas in pregnancy-induced hypertension syndrome

Objective To examine the expression of the hypoxia-ind uc ible factor 2α(HIF-2α)in placentas from women with normal pregnancies or pre eclampsia,and investigate the relationship between HIF-2α and the etiology of preeclampsia.Methods Placentas were obtained from ten women wit h preeclampsia and ten gestational age-matched women with normal pregnancies un dergoing cesarean section without labor at term.The HIF-2α expression of prot ein was detected by western-blotting;the HIF-2α expression of mRNA was detect ed by real-time PCR;the HIF-2α localizationofproteinin placentasfrompreeclampsiabyimmunohistochemistry .Results The protein and mRNA l evels of HIF-2α in placentas with preeclampsia were significantly higher than those with normal pregnancies.The HIF-2α immunoreactivity was mainly located i n the nuclei of the syncytiotrophoblast and fetoplacental vascular endothelium i n the preeclamptic placentas.Conclusion The overexpression of H IF-2α in placentas with preeclampsia confirms the hypothesis of placental hypo xia,it may play an important role in the pathogeny and pathophysiologic process of preeclampsia.
