雷帕霉素对糖尿病肾病大鼠肾脏保护作用的实验研究

摘    要:目的探讨雷帕霉素对糖尿病肾病大鼠肾脏的保护作用。方法将24只雄性SD大鼠随机分为正常对照组(C组)、糖尿病肾病模型组(DN组)和雷帕霉素治疗组(R组)各8只。R组大鼠给予雷帕霉素灌胃。观察各组大鼠血糖、24h尿蛋白及肾脏病理改变,并通过透射电镜观察足细胞的超微结构的改变。结果与C组相比,DN组大鼠尿蛋白排泄量明显增加(P<0.01),肾脏病理可见肾小球肥大,电镜下可见肾小球足细胞足突增宽、融合,基底膜节段性增厚;R组大鼠尿蛋白较DN组降低(P<0.01),肾组织病理改变减轻。结论雷帕霉素对糖尿病肾病具有一定肾脏保护作用,可能通过改善足细胞超微结构,延缓糖尿病肾病的进展。

Abstract:Objective To investigate the protective effects of Rapamycin on kidney in diabetic nephropathy (DN) rats. Methods Twenty-four SD male rats were randomly divided into normal control group (group C, n=8), DN group (group DN, n=8), and treated group with Rapamycin (group R, n=8). Rats in Group R were lavaged with Rapamycin; Group C and Group DN were lavaged with 0.5% carboxymethyl cellulose (CMC) at the same volume. Measure changes in blood glucose, 24-hour urine protein. Pathological changes of glomerulus were tested by HE staining, and ultra microstructure of glomerulus by electron microscope. Results Compared with Group C, Group DN's blood glucose, 24-hour urinary protein had significantly risen (P〈0. 01) kidneys were enlarged, podocyte foot process widened and partially in fusion; Group R's 24-hour urinary protein were significantly lower than Group DN (P〈0.01), and glomerular fibrosis was reduced significantly in Rapamycin treatment group. Conclusion Rapamycin may be play a role in protection in rats with DN, it maintains the integrity of podocyte structure, and further delayed the progress of DN.
