基于UPLC-Q-TOF-MS/MS技术的彝族药姜味草化学成分分析

摘    要:目的 应用超高效液相色谱-串联四级杆飞行时间质谱联用技术(UPLC-Q-TOF-MS/MS)对彝族药姜味草中的化学成分进行定性分析。方法 Waters Acquity UPLC BEH C18色谱柱(2.1 mm×100 mm,1.7 μm),以乙腈(A)-体积比0.1%甲酸水溶液(B)梯度洗脱,质谱分析采用信息关联模式(IDA),在正、负离子模式下采集数据。结果 根据化合物的精确分子质量和碎片离子信息,结合对照品比对和文献数据,从姜味草中鉴定推断出77个化学成分,包括黄酮类27个、有机酸类21个、苯丙素类8个、三萜酸类8个、氨基酸类6个,其他类:蒽醌类、环烯醚萜类、醇类等7个。结论 首次对姜味草中的化学成分进行快速鉴别,为其质量控制及药效物质基础提供了科学依据。

Abstract:OBJECTIVE To analyze and identify the chemical constituents from Yi medicine Micromeria biflora (Buch.-Ham.exD.Don) Benth. by using UPLC-Q-TOF/MS. METHODS The separation was performed on Waters Acquity UPLC BEH C18(2.1 mm×100 mm,1.7 μm) column with gradient elution of acetonitrile(A)-0.1% formic acid(B). The MS analysis was based on information associated mode(IDA), and positive and negative ions were collected respectively. RESULTS Fragments were analyzed and identified according to the precise mass charge ratio of the compounds, secondary mass spectrometry, and comparison of reference materials and literature data. A total of 77 compounds were identified, including 27 flavonoids, 21 organic acids, 8 phenylpropanoids, 8 triterpenoids, 6 amino acids, 7 anthraquinones, iridoids and alcohols. CONCLUSION The chemical components of Micromeria biflora (Buch-Ham.ex D.Don) Benth. are rapidly identified for the first time, which lays a foundation for the research of quality control and pharmacodynamic material basis.
