小鼠赖氨酰氧化酶样2基因的组织特异性表达

摘    要:[目的 ]研究小鼠赖氨酰氧化酶样 2基因 (mLOXL2 )的组织特异性表达 .[方法 ]应用Blast检查程序、聚合酶链反应、Northern印迹法和免疫组织化学染色法 .[结果 ]mLOXL 2cDNA推断的氨基酸序列呈现赖氨酰氧化酶 (LOX)家族的特征性区域 ,即铜结合部位和催化区域 .进化距离指出小鼠赖氨酰氧化酶 (mLOX)家族有 2个独特的组 ,第 1组只包含LOX基因 ,第 2系统发育组包含所有其它赖氨酰氧化酶样基因 (LOXL) .mLOXL 2在肺及甲状腺中有高表达 ,表皮、毛发滤泡周围和骨骼肌内有LOXL 2的强染色 ,软骨细胞内有LOXL 2的胞质染色 .[结论 ]鼠类的氨基酸序列显示和人类mLOXL2cDNA一致的高水平的同族关系 ,在几乎所有组织中mLOXL 2基因被表达为 4kbmRNA ,并在肺和胸腺中有高度稳定状态mRNA水平 .人类和鼠类组织中 ,LOXL 2将在控制分子和细胞相互作用上起重要的作用 .

Abstract:OBJECTIVETo study the tissue-specific expression of the mouse lysyl oxidase like-2 gene (mLOXL2).METHODSIt was used that the Blast search program, polymerase chain reaction (PCR), Northern blot and immunohistochemistry.RESULTSThe deduced amino sequence of mLOXL2 cDNA presented the characteristic domains of lysyl oxidase (LOX) family, the copper-binding site and the catalytic domain. The evolutionary distance indicated two distinct groups; the first group included the LOX gene only and the second phylogenic group included all the other lysyl oxidase-like genes(LOXL). The mLOXL2 gene had a high expression in lungs and thymus. A strong staining for LOXL2 was detected in epidermis, around the hair follicle and in the skeletal muscle and cartilage cells had cytoplasmic staining for LOXL2.CONCLUSIONA mouse cDNA encoding a protein that shares similarities with the previously characterized mLOX has been cloned and sequenced, and the mLOXL2 gene is expressed as a 4kb mRNA in almost all tissues and the highest steady state mRNA levels were found in lung and thymus. The LOXL2 will play an important role or will give a high importance in the ability to control molecular and cellular interaction.
