HPLC法测定肿节风中绿原酸、异嗪皮啶和迷迭香酸的含量

目的建立同时测定肿节风药材中绿原酸、异嗪皮啶及迷迭香酸含量的高效液相色谱法。方法采用Phenomenex Gemini C18柱(250mm×4.6mm,5μm)分离,以乙腈和4mL·L-1磷酸水溶液进行梯度洗脱,流速1.0mL·min-1,检测波长327nm。结果绿原酸在4.48⁴4.8mg·L-1范围内线性关系良好,平均回收率为101.5%(RSD为2.1%),异嗪皮啶在1.7244.8mg·L-1范围内线性关系良好,平均回收率为101.5%(RSD为2.1%),异嗪皮啶在1.72¹7.2mg·L-1范围内线性关系良好,平均回收率为97.7%(RSD为2.7%),迷迭香酸在1.8617.2mg·L-1范围内线性关系良好,平均回收率为97.7%(RSD为2.7%),迷迭香酸在1.86¹8.6mg·L-1范围内线性关系良好,平均回收率为99.3%(RSD为2.4%)。结论含量测定方法操作简便,准确度高,重复性好,为肿节风药材质量控制和评价提供了可靠的方法。

Determination of chlorogenic acid,isof raxidin and rosmarinic acid in Sarcand ra glabra by HPLC

Objective To develop an HPLC method for determining chlorogenic acid ,isofraxidin and rosmarinic acid in Sarcandra glabra .Methods The analysis was carried out with a Phenomenex Gemini C18 column（250 mm ×4 .6 mm ,5 μm） .The mobile phase was composed of acetonitrile-4 mL · L -1 phosphoric with gradient elution ,at a flow rate of 1 .0 mL ·min-1 .The detecting wavelength was at 327 nm .Results Chlorogenic acid ,isofraxidin and rosmarinic acid were linear in the range of 4 .48-44 .8 ,1 .72-17 .2 and 1 .86-18 .6 mg ·L -1 .The average recoveries were 101 .5% （RSD 2 .1% ） ,97 .7% （RSD 2 .7% ） ,99 .3% （RSD 2 .4% ） ,re-spectively .Conclusion The method is quick ,simple ,and reproducible ,which could be used to control the quality of Sarcandra gla-bra .
