摘    要:目的建立以HPLC法同时测定人血浆中甘草酸和苦参碱质量浓度的方法。方法 Ultimate AQ-C18柱(250mm×4.6mm,5μm),柱温为室温(25℃),流动相为乙腈-三乙胺调pH至7.5的0.05mol·L-1磷酸二氢钾(22.5∶77.5),流速为1.0mL·min-1,检测波长为220nm(检测苦参碱)和250nm(检测甘草酸),进样量50μL。结果甘草酸单铵盐质量浓度在2.0200.0μg·mL-1范围内线性良好,苦参碱质量浓度在2.0200.0μg·mL-1范围内线性良好,苦参碱质量浓度在2.0200.0μg·mL-1范围内线性良好。平均回收率均大于95%,日内、日间RSD均小于4%。结论该方法简便、快速、灵敏、可靠,可用于人血浆中甘草酸和苦参碱质量浓度的监测。

Abstract:Objective To establish an HPLC methods for the simultaneous determination of human plasma concentration of glycyr-rhizic acid and matrine .Methods Ultimate AQ-C18 (250 mm × 4 .6 mm ,5 μm) was chosen as the chromatographic column .The column temperature was at room temperature(25 ℃) .The mobile phase was composed of acetonitrile and 0 .05 mol · L -1 aqueous phosphoric acid(22 .5∶77 .5 ,adjusted to pH 7 .5 with triethylamine) .The flow rate was set at 1 .0 mL · min-1 .The detection wavelengths were set at 220 and 250 nm .The injection volume was 50μL .Results The linear ranges of glycyrrhizic acid ammoni-um salt and matrine were 2 .0-200 .0 μg · mL -1 and 2 .0-200 .0 μg · mL -1 ,respectively .The average recovery was above 95% ;both the intraday RSD and the interday RSD were less than 4% .Conclusions The method is simple ,rapid ,sensitive and reliable , and could be used for the simultaneous determination of human plasma concentration of glycyrrhizic acid and matrine .
