长双歧杆菌、肠系膜明串珠菌和鼠李糖乳杆菌对小鼠血清超氧化物歧化酶、谷胱甘肽过氧化物酶、丙二醛水平及肝细胞脂肪变性的影响

摘    要:目的探讨长双歧杆菌(BL)、肠系膜明串珠菌(LM)和鼠李糖乳杆菌(LR)对小鼠血清抗氧化活性的促进作用及对小鼠肝细胞脂肪变性的影响。方法雄性健康清洁级Balb/c小鼠50只,随机分为对照组、模型组、BL干预组、LM干预组及LR干预组,每组10只。模型组、BL干预组、LM干预组、LR干预组小鼠每日饲以高脂饲料,连续15周,并同时分别予以生理盐水或BL、LM、LR灌胃,连续15周,检测小鼠血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)水平,并观察细胞脂肪变性。结果对照组小鼠无肝脂肪变性8只(80%),轻度脂肪变性2只(20%);模型组小鼠呈现中、重度肝脂肪变性分别为4只(40%)、6只(60%);BL干预组中、重度脂肪变性分别为5只(50%)、4只(40%),LM干预组中、重度肝脂肪变性均为4只(分别占40%),与对照组比较差异有统计学意义(P<0.05),而与模型组比较差异无统计学意义(P>0.05);LR干预组小鼠以轻度肝脂肪变性为主,共8只(80%),与模型组比较差异有统计学意义(P<0.05)。与对照组比较,模型组小鼠SOD和GSH-Px活性显著降低(P<0.05),MDA水平显著升高(P<0.05);与模型组比较,BL干预组和LM干预组小鼠SOD和GSH-Px活性、MDA水平差异无统计学意义(P>0.05),而LR干预组SOD和GSH-Px活性显著升高(P<0.05),MDA水平显著降低(P<0.05);BL干预组与LM干预组小鼠比较SOD和GSH-Px活性、MDA水平差异无统计学意义(P>0.05),而LR干预组小鼠SOD和GSH-Px活性较BL干预组、LM干预组均显著升高(P<0.05),MDA水平较BL干预组、LM干预组均显著降低(P<0.05)。与对照组比较,模型组小鼠肝功能指标及肝指数均显著升高(P<0.05);与模型组比较,BL干预组和LM干预组小鼠肝功能指标及肝指数差异无统计学意义(P>0.05),而LR干预组除总胆固醇(TC)外,三酰甘油(TG)、丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)和肝指数均显著降低(P<0.05);BL干预组和LM干预组小鼠肝功能指标及肝指数差异无统计学意义(P>0.05);而LR干预组小鼠除TC外,TG、ALT、AST和肝指数均较BL干预组和LM干预组显著降低(P<0.05)。结论 LR对小鼠血清的抗氧化活性有加强作用,显著抑制了小鼠肝细胞脂肪肝变性。

Abstract:Objective To investigate the effect of application of Bifidobacterium longum (BL), Leuconostoc mesenteroides(LM) and Lactobacillus rhamnosuson (LR)on serum antioxidant activity enhancement and liver steatosis inhibition of mice. Methods Fifty mice of male health clean level were randonrly divided into five groups including control group, model group, BL group, LM group and LR group with 10 mice in each group. The model, BL, LM and LR groups were daily oral administrated with high-fat diet and physiological saline, BL, LM and LR respectively for 15 weeks. The serum superoxide dismutase ( SOD), glutathione peroxidase (GSH-Px) activity and malondialdehyde (MDA) level were tested and the steatosis ceils were observed. Results Eight mice (80%) had no fatty degeneration and two mice (20%) had mild fatty degeneration in control group. The number of mice who had moderate and severe fatty degeneration of the liver was four cases(40% ) and six cases (60%)in model group,five cases(50% ) and four cases(40% ) in BL group and both was four cases(40% ) in LM group,which had statistically significant differences of BL and LM group compared with the control group ( P 〈 0.05 ), but had no sta- tistical significance compared with the model group(P 〉 0.05 ). There were eight cases(80% ) with mild hepatic steatosis in LR group,which had statistically significant difference compared with model group (P 〈 0.05 ). Compared with control group, SOD and GSH-Px activity decreased significantly while MDA increased significantly in model group ( P 〈 0.05 ) ; compared with model group, SOD and GSH-Px activity and MDA level in BL group and LM group had no significant difference(P 〉 0.05 ), while the SOD and GSI-I-Px activity increased significantly and the MDA level decreased significantly in LR group (P 〈 0.05 ) ; there was no statistical significance of SOD and GSH-Px activity and MDA level between BL group and LM group(P 〉 0.05 ), while SOD and
