多巴胺能药物治疗帕金森病的长时程反应

长期以来左旋多巴为帕金森病（PD）药物治疗的金标准。但众所周知,PD随左旋多巴治疗时间延长,会不可避免地出现运动症状并发症,此现象与多巴制剂的长时程反应（LDR）逐渐消退密切关联。由此,探究和恰当掌握抗PD药物的长时程反应特性,不仅有助于深入研究PD的病理机制,也有益于合理药物治疗方案的运用,最大化发挥抗PD药物作用效能。本研究就LDR的形成时机、持续时间及强度等表现特征,以及形成机制、临床意义等方面进行阐述,以期能为PD临床治疗策略做出更好选择。

The Long-duration Response of Dopaminergic Therapy in Parkinson＇s Disease

Levodopa has been regarded as a gold standard symptomatic treatment for Parkinson＇s disease（PD）.As the duration of administration prolonging,however,levodopa would lead inevitably to motor complications,which is believed to be related to the fading of the long-duration response（LDR） of the dopaminergic therapy.To study the response-characteristics of dopaminergic medications will benefit not only to exploring PD pathogenesis,but also to optimizing therapeutic regimen in the treatment.The LDR phenomenology about its occurrence,duration,magnitude,potential mechanisms and clinical significance were discussed.
