海螵蛸作为组织工程支架材料的实验研究

摘    要:目的了解并观察海螵蛸的空间结构及作为组织工程支架的生物相容性。方法将脱脂海螵蛸切成薄片,计算孔隙率,从光学显微镜和扫描电镜观察其空间结构。在小鼠背部皮下分别埋置海螵蛸支架和聚乙酸醇(PGA),在术后1、2、4、8周观察炎症反应和吸收情况。四甲基偶氮唑蓝比色法(MTT assay)分别检测海螵蛸支架对细胞增殖功能的影响。结果网格状的海螵蛸具有92%得孔率。皮下埋植术后1、2周仅少许淋巴细胞浸润,4周部分吸收,8周完全吸收,炎症细胞基本消退,与PGA比较无统计学差异。MTT assay测得细胞在海螵蛸支架中增殖能力较强。结论从结构上观察,海螵蛸符合细胞培养支架的多孔隙要求,且具有良好的生物相容性。

Abstract:Purpose To observe the spatial structure of cuttlebone and to study the biocompatibility of cuttlebone used as scaffolds in tissue engineering. Methods The degreased cuttlebone was examined with light microscope and scanning electron microscope. The porosity rate was calculated. The cuttlebone and polyglycolic acid (PGA) were placed subcutaneously in the mice back. The inflammatory reaction and absorbing degree of cuttlebone were investigated at 1, 2, 4 and 8 weeks post-operation. The effect of cuttlebone on cell proliferation was examined by MTT assay. Results Morphological study showed regular network. The porosity rate was 92%. Only a few lymphocytes were infiltrated after 1 to 2 weeks. Cuttlebone was partially absorbed after 4 weeks and fully degraded after 8 weeks. Cell proliferated well on the cuttlebone scaffold. Conclusions Cuttlebone exhibits satisfactory biocompatibility and can be used as suitable three-dimensional porous scaffold.
