寻常性银屑病患者外周血单一核细胞TLR2,TLR4,TLR9表达的初步研究

摘    要:目的研究寻常性银屑病患者外周血单一核细胞(PBMCs)Toll样受体2(TLR2) mRNA,TLR4 mRNA和TLR9 mRNA的表达情况。方法采用实时荧光定量PCR方法检测30例寻常性银屑病患者(其中发病与感染相关者10例,与感染无关者20例)和25例正常人PBMCs内TLR2,TLR4,TLR9 mRNA的水平。结果与感染有关银屑病组、与感染无关银屑病组较正常对照组TLR9 mRNA水平差异有显著性(P值分别为0.036和0.016),与感染无关银屑病组与正常对照组比较差异无显著性(P>0.05);TLR2和TLR4的水平在三组之间差异无显著性(P>0.05)。结论TLR9的表达在与感染相关银屑病患者中显著升高,提示感染因素,特别是细菌CpGDNA在寻常性银屑病中的作用可能通过TLR9介导。

Abstract:Objective To investigate the expression of TLR2,TLR4 and TLR9 mRNA in peripheral blood mononuclear cells (PBMC) of patients with psoriasis vulgaris.Methods Total RNA from patients and healthy controls were extracted and TLR2,TLR4 and TLR9 mRNA expression were detected by real-time quantitative PCR.Results TLR9 mRNA level was significantly up regulated in the patients with infection compared to that of patients without infection (P=0.036) and controls (P=0.016).However,there was no significant difference in expression levels of TLR2 and TLR4 mRNA among the patients with or without infection and healthy controls (P>0.05).Conclusion The high expression of TLR9 in the psoriatic patients associated with infecation indicate that bacterial CpG DNA may also play a role in the pathogenesis of psoriasis.
