加味温胆汤对抑郁模型大鼠海马CaMK信号转导通路的影响

摘    要:目的 探讨加味温胆汤对抑郁模型大鼠海马钙调素依赖蛋白激酶(CaMK)信号转导通路的影响.方法 128只SD大鼠随机分为空白组、模型组、中药组、西药组,每组32只.采用孤养结合慢性不可预见性应激造模抑郁大鼠,从造模第1天开始中药组给予加味温胆汤12g/(kg·d),西药组予氟西汀1.8mg/(kg·d),模型组、空白组给予生理盐水2ml/(kg·d),各组均灌胃给药,连续28天.实验第7、14、21、28天时采用Western印迹法检测各组大鼠海马N-甲基-D-天冬氨酸受体1(NMDA-NR1)、钙调素(CaM)、CaMKⅡ蛋白表达变化.结果 第7、14天,各组大鼠海马NMDA-NR1、CaM、CaMKⅡ蛋白表达差异无统计学意义(P>0.05).21、28天时,与空白组比较,模型组大鼠海马NMDA-NR1、CaM、CaMKⅡ蛋白表达增多(P<0.01);与模型组比较,中、西药组大鼠海马NMDA-NR1、CaM、CaMKⅡ蛋白表达减少(P<0.05或P<0.01);中、西药组同时间比较,差异无统计学意义(P>0.05).结论 加味温胆汤可通过抑制CaMK信号转导通路的活性发挥其抗抑郁效应.

Abstract:Objective To observe the effect of Jiawei Wendan Deco ction(Supplemente d Gallbladder-Warming Decoction) on calmodulin-dependent protein kinase(CaMK) s ignaling pathway in the hippocampus of depression model rats.Methods The rat models w ere established with unpredictable chronic stress and isolation-induced depress ion.Then 128 models were randomized into blank group,model group,Chinese m edication group and western medication group,with 32 in each.The Chinese medic ation group and western medication group was given 12g/(kg·d) of Jiawei Wendan Decoction and 1.8mg/(kg·d) of fluoxetine through gastric feeding respectively. Both the model group and blank group were given 2ml/(kg·d) of normal saline thro ugh gastric feeding.The administration lasted for 28 days. On the 7th,14th,21st and 28th day,the protein expression of N-methyl-D-aspartate receptor 1(NMDA-NR1),calmodulin(CaM) and CaMKⅡ in the hippocampus of rat models were tested with Western-Blot technique.Results The re was no statistical significance in the protein expression of NMDA-NR1,CaM a nd CaMKⅡamong groups on the 7th and 14th day of administration(P>0.05).On the 21st and 28th day,the protein expression of NMDA-NR1,CaM and CaMKⅡ increased significantly in the model group as compared with the blank group(P<0.01). The protein expression of NMDA-NR1,CaM and CaMKⅡ decreased significantly in the t wo medication groups as compared with the model group(P<0.05 or P<0.01).The differen ce between two medication groups was not statistically significant at the same time p eriod(P>0.05).Conclusion Jiawei Wendan Decoction has an antidepressant effect by inhibiting the activity of CaMK signaling pathway.
