摘    要:目的 探讨miR-638在人骨肉瘤(OS)细胞增殖行为中的影响。方法通过实时定量聚合酶链反应(qRT-PCR)检测miRNA-638在人OS组织及癌旁组织、人OS细胞株(MG63、U2OS、HOS、SaOS2)和正常人成骨NHOst细胞株中的表达;用LipofectamineTM 2000转染miR-638模拟物或抑制物,细胞增殖检测试剂盒(CCK-8)检测转染后的吸光光度值,平板克隆形成实验检测菌落数量。结果miR-638在各组细胞中均有表达;OS细胞株中miR-638的表达明显下调(P<0.05);高表达miR-638能抑制OS MG63和U2OS细胞的增殖(P<0.05)。 结论miR-638高表达能抑制OS细胞的增殖。

Abstract:Objective To investigate the role of miR-638 in biological behavior of osteosarcoma cell.MethodsThe expression of miR-638 in osteosarcoma MG63 and U2OS cells was detected by RT-PCR.miRNA-638 mimics (inhibhors) were transfected into MG63 and U2OS by using LipofectamineTM2000 to upregulate(deregulate) the expression of miRNA-638.The absorbance value in 24 h,48 h,96 h and 72 h following the transfection was detected by CCK-8.And colony forming assay was used to detect colony counts over 30 cells after 2 weeks.ResultsMiR-638 expressed in each group.The expression of miR-638 was down regulated (P<0.001) in the 36 groups (56.25%) compared with the corresponding non cancerous tissues.Mir-638 expression can significantly reduce the MG63 and U2OS cells growth rate (P<0.05),on the contrary,the suppression of miR-638 expression significantly promoted the growth of cells of U2OS and MG63 (P<0.05).ConclusionsHigh expression of miR-638 significantly inhibits the proliferation of osteosarcoma cells.MiR-638 may become a new target for the early diagnosis and treatment of osteosarcoma.
