酒精对幼鼠神经行为发育的影响

摘    要:目的 探讨酒精对雌性幼鼠神经行为发育的影响。方法  4 8只 8周龄昆明雌性幼鼠随机分成对照组、低、中和高浓度组 ,分别用不同浓度的酒精 (0 %、12 5 %、2 5 %和 5 0 % )灌喂 ,按 10ml/kg ,隔天 1次 ,连续 90d ,采用自发活动试验、Morris水迷宫和免疫组化等技术 ,结合图像分析系统定量分析酒精对神经行为与大脑形态变化相关性。结果 ①低浓度组自发活动增加 ,高浓度组自发活动减少 ,高浓度组的Morris水迷宫潜学习成绩延长 :②高浓度组脑重量减轻 ,大脑内侧中央前区(Prcm)、第 1感觉运动皮质区 (S1)和无粒型岛皮质后区 (Alp)、纹状皮质 18区 (Str18)和颞叶听区 (TeAud)皮质厚度明显变薄 ;③高、中浓度组胶质细胞纤维酸蛋白 (GFAP)免疫反应强度增强及反应性胶质细胞的平均相对密度升高。结论 高、中浓度酒精对小鼠神经行为发育有抑制作用。

Abstract:Objective To investigate the effects of alcohol exposure on neurobehavioral development of immature female mouse. Methods Forty-eight immature mice were divided randomly into the control group and the low, middle and high dose groups. Different doses ethanol (0%,12.5%, 25% and 50%) was given (10 ml/kg) intragastrically by gavages once every other day for 90 days respectively. The relationship of cerebral structure changes and neurobehavior was analyzed in fixed quantity by using spontaneous activity, Morris water labyrinth and immunohistochemistry, combining with pathologic image analysis system. Results ① The low doses group increase their activities obviously in the spontaneous activity; on the contrary, the high group decrease their activities obviously (P<0.05). The latency was prolonged significantly in the high doses groups in Morris water labyrinth (P<0.05).② The weight of brain in high doses group was lightened. The cerebral cortex thickness in the areas of Prcm, Sl,Str18,Alp and TeAud was reduced obviously. ③ Enhancement of immunity reaction in glial fibrillary acidic protein and increase of average relative density in reacting glial fibrillary were significant in both high and middle doses groups. Conclusion The middle and high doses alcohol can damage neurobehavioral development of immature mouse.
